Our &#8220;rest-and-digest&#8221; response requires it. Acetylcholine also plays an important role in learning, memory, and <a href=\"https:\/\/selfhacked.com\/blog\/health-benefits-sleep\/\">sleep<\/a> quality. So what happens when acetylcholine runs low? In this post, we&#8217;ll explore the science to see if there such as thing as &#8220;acetylcholine deficiency&#8221; and whether certain lifestyle changes and nutrients can increase acetylcholine levels.  <\/p>\n\n\n\n<!--more-->\n\n\n\n<h2><span id=\"Does_Acetylcholine_Deficiency_Exist\"><strong>Does &#8220;Acetylcholine Deficiency&#8221; Exist?<\/strong><\/span><\/h2>\n\n\n\n<h3><span id=\"Research_Limitations\"><strong>Research Limitations<\/strong><\/span><\/h3>\n\n\n\n<p>Due to its wide range of roles throughout the body and brain, low acetylcholine levels or activity in certain brain areas have been implicated in the development, progression, or symptoms of a variety of health conditions.<\/p>\n\n\n\n<p>However, most of the studies dealt with associations&nbsp;only, which means that a cause-and-effect relationship hasn\u2019t been established.<\/p>\n\n\n\n<p>For example, just because dementia has been linked with low acetylcholine activity in certain brain areas doesn\u2019t mean that cognitive dysfunction is always caused by &#8220;acetylcholine deficiency.&#8221; Data are lacking to make such claims.&nbsp;<\/p>\n\n\n\n<p>Also, even if a study did find that low acetylcholine contributes to cognitive dysfunction, acetylcholine levels are highly unlikely to be the only cause. Complex disorders like dementia always involve multiple possible factors \u2013 including brain chemistry, environment, health status, and genetics \u2013 that may vary from one person to another.<\/p>\n\n\n\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Acetylcholine is undoubtedly an important neurotransmitter, but whether low acetylcholine levels in specific brain areas can <em>cause<\/em> health problems is still uncertain.<\/div><\/div>\n\n\n\n<h3><span id=\"Associated_Conditions\"><strong>Associated Conditions:<\/strong><\/span><\/h3>\n\n\n\n<p><strong><em>There are no symptoms associated with &#8220;acetylcholine deficiency&#8221; or low acetylcholine levels per se.<\/em><\/strong><em>&nbsp;<\/em> <em>Instead, people may only show symptoms of mental health or neurocognitive disorders. Your doctor will discuss your symptoms with you and run tests to pinpoint the underlying cause.<\/em> <\/p>\n\n\n\n<p><em>Low or high acetylcholine levels don\u2019t necessarily indicate a problem if there are no symptoms or if your doctor tells you not to worry about it.<\/em><\/p>\n\n\n\n<h3><span id=\"1_Dementia\">1) <strong>Dementia<\/strong><\/span><\/h3>\n\n\n\n<p>Some studies suggest that poor cholinergic activity and low acetylcholine in the brain may cause cognitive decline and poor memory in people with dementia. Drugs used to slow down dementia work precisely by increasing acetylcholine in the brain [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4865532\/\">R<\/a>].<\/p>\n\n\n\n<h3><span id=\"2_Alzheimers_Disease\">2) Alzheimer&#8217;s Disease<\/span><\/h3>\n\n\n\n<p>Acetylcholine has also been proposed to play a potentially significant role in the development or symptoms of certain common neurodegenerative diseases, including <a href=\"https:\/\/selfhacked.com\/blog\/alzheimer\/\">Alzheimer\u2019s disease<\/a> [<a href=\"http:\/\/webspace.ship.edu\/cgboer\/genpsyneurotransmitters.html\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2654795\/\">R<\/a>].<\/p>\n\n\n\n<p>In Alzheimer\u2019s disease, the so-called \u201c<em>cholinergic<\/em>\u201d neurons \u2014 the brain cells that primarily use acetylcholine \u2014 gradually become damaged and destroyed. Additionally, important molecules called <em>acetylcholine transporters<\/em> may also become impaired as the disease progresses. These molecules are responsible for transporting acetylcholine into neurons: and impairing them can, therefore, make it more difficult for acetylcholine to fulfill its normal functions throughout the brain. Together, then, these two important acetylcholine-related mechanisms are likely to contribute significantly to some of the hallmark cognitive symptoms of these disorders [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24590577\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3243760\/\">R<\/a>].<\/p>\n\n\n\n<p>Many (though not all) drugs that are currently used to treat Alzheimer\u2019s disease are <a href=\"https:\/\/selfhacked.com\/blog\/alzheimer\/#1_Acetylcholinesterase_Inhibitors\"><em>acetylcholinesterase inhibitors<\/em><\/a>. As their name suggests, these drugs inhibit the enzyme <em>acetylcholinesterase<\/em>, which is responsible for breaking down the neurotransmitter acetylcholine throughout the brain. Therefore, inhibiting these enzymes can result in an overall increase of acetylcholine levels and activity, which may potentially compensate for the loss of cholinergic neurons that typically occurs in Alzheimer\u2019s disease [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24590577\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3243760\/\">R<\/a>].<\/p>\n\n\n\n<p>For example, two common Alzheimer\u2019s medications, <a href=\"https:\/\/selfhacked.com\/blog\/galantamine-12-proven-health-benefits-galantamine-backed-science\/\"><em>galantamine<\/em><\/a> and <a href=\"https:\/\/selfhacked.com\/blog\/alzheimer\/#1_Acetylcholinesterase_Inhibitors\"><em>donepezil<\/em><\/a>, are each acetylcholinesterase inhibitors \u2014 and their therapeutic effects in relieving some of the cognitive symptoms of Alzheimer\u2019s are believed to stem primarily from to their ability to stimulate acetylcholine activity throughout the brain [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/12962529\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/12137632\">R<\/a>, <a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/26942549\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/29923184\">R<\/a>].<\/p>\n\n\n\n<p>However, not all Alzheimer\u2019s medications target acetylcholine. For example, the widely-used Alzheimer\u2019s drug <a href=\"https:\/\/selfhacked.com\/blog\/memantine\/\"><em>memantine<\/em><\/a> targets other mechanisms entirely (specifically, NMDA receptors).<\/p>\n\n\n\n<p>Therefore, acetylcholine is probably only one piece of a much more complex puzzle. Nonetheless, it does appear to play an important role in at least some of the main mechanisms and symptoms of Alzheimer\u2019s disease \u2014 and more research will be needed to explore these mechanisms more fully, as well as to potentially develop future medical treatments for these neurodegenerative disorders.<\/p>\n\n\n\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Alzheimer&#8217;s disease is marked by a loss of neurons that produce acetylcholine. Drugs that increase acetylcholine levels are commonly prescribed to manage the cognitive symptoms of Alzheimer&#8217;s.<\/div><\/div>\n\n\n\n<h3><span id=\"3_Parkinsons_Disease\">3) Parkinson&#8217;s Disease<\/span><\/h3>\n\n\n<p>According to limited studies, people with Parkinson\u2019s disease may also have low acetylcholine levels in the brain [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/14676050\">R<\/a>].<\/p>\n<p>On the other hand, low acetylcholine levels might be a consequence, not a cause, of these neurodegenerative diseases. More research is needed [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/14676050\">R<\/a>].<\/p>\n<h3><span id=\"4_Myasthenia_Gravis\"><strong>4) Myasthenia Gravis<\/strong><\/span><\/h3>\n<p>Myasthenia Gravis is an autoimmune disease where acetylcholine nerves that go to the muscles become destroyed, causing severe muscle weakness. Future work should clarify this association [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/15888431\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2687934\/#R80\">R<\/a>].<\/p>\n<h3><span id=\"6_Glaucoma\">6) <strong>Glaucoma<\/strong><\/span><\/h3>\n<p>Acetylcholine helps maintain healthy vision and eye blood pressure. Anticholinergic drugs often prescribed to treat overactive bladder can trigger glaucoma [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/30120418\">R<\/a>].<\/p>\n<h3><span id=\"7_Autism\">7) <strong>Autism<\/strong><\/span><\/h3>\n<p>According to one controversial theory, children with autism may lack brain acetylcholine, which can contribute to intellectual impairment and epilepsy. Theoretically, acetylcholine can increase the \u201ccalming\u201d neurotransmitter <a href=\"https:\/\/selfhacked.com\/blog\/natural-ways-to-increase-gaba\/\">GABA<\/a>, which reduces the chance of seizures [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/20190638\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24096295\">R<\/a>].<\/p>\n<p>In mice with autism, increasing acetylcholine improved cognitive and social symptoms. However, this link hasn\u2019t been proven in humans [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/20190638\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24096295\">R<\/a>].<\/p>\n<h3><span id=\"Can_Acetylcholine_Deficiency_Be_Confirmed\">Can &#8220;Acetylcholine Deficiency&#8221; Be Confirmed?<\/span><\/h3>\n<p>Although low acetylcholine levels are commonly described as \u201cacetylcholine deficiency,\u201d this can be misleading. Assessing acetylcholine levels in the brain is extremely difficult.<\/p>\n<p>For one, no deficiency parameters have been established in the scientific or medical literature.<\/p>\n<p>Secondly, acetylcholine levels can\u2019t be directly measured. Even with indirect measurements (such as with brain imaging), acetylcholine levels may vary across brain areas and quickly change depending on many factors.<\/p>\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Currently, no direct-to-consumer test can assess acetylcholine levels. Brain imaging can estimate levels in research settings, but no definition of &#8220;acetylcholine deficiency&#8221; has been widely accepted.<\/div><\/div>\n\n\n<h2><span id=\"What_Influences_Acetylcholine_Levels\"><strong>What Influences Acetylcholine Levels<\/strong>?<\/span><\/h2>\n\n\n\n<h3><span id=\"Acetylcholine_Signaling_Its_Not_That_Simple\">Acetylcholine Signaling: It&#8217;s Not That Simple<\/span><\/h3>\n\n\n\n<p>Many biological processes and pathways are involved in determining the total amount of acetylcholine in the body and brain, as well as its overall degree of activity.<\/p>\n\n\n\n<p><strong>This means that there are many different mechanisms and pathways that can influence acetylcholine, such as:<\/strong><\/p>\n\n\n\n<ul><li>Increasing or decreasing the levels of its \u201cingredients\u201d (<em>metabolic precursors<\/em>), such as <a href=\"https:\/\/selfhacked.com\/blog\/top-18-scientific-health-benefits-choline-cdp-choline-alpha-gpc\/\"><em>choline<\/em><\/a><\/li><li>Activating or inhibiting the enzymes that produce (<em>synthesize<\/em>) active acetylcholine from its precursors, such as <a href=\"https:\/\/selfdecode.com\/gene\/CHAT\/?utm_source=seo&amp;utm_medium=selfhacked&amp;utm_campaign=id00002\"><em>choline acetyltransferase<\/em><\/a> or <em>acetyl-coenzyme A<\/em><\/li><li>Stimulating or suppressing the release of acetylcholine by nervous system cells<\/li><li>Directly activating acetylcholine receptors, such as by \u201cimitating\u201d natural (\u201c<em>endogenous<\/em>\u201d) acetylcholine<\/li><li>Blocking acetylcholine receptors, thereby preventing them from being activated by natural acetylcholine<\/li><li>Increasing or decreasing the number of acetylcholine receptors<\/li><\/ul>\n\n\n\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Acetylcholine balance is complex because it can be influenced by building blocks, other neurotransmitters, feedback loops, different brain areas, and many receptors and enzymes.<\/div><\/div>\n\n\n\n<h3><span id=\"Why_Target_The_Acetylcholine_System\"><strong>Why Target The Acetylcholine System?<\/strong><\/span><\/h3>\n\n\n\n<p>Because acetylcholine is believed to be involved in the development of a variety of diseases and other health conditions, there are many <a href=\"https:\/\/selfhacked.com\/blog\/anticholinergics\/\">potential uses<\/a> that have been proposed for substances that can target the acetylcholine system.<\/p>\n\n\n\n<p>Additionally, many people in the \u201c<a href=\"https:\/\/selfhacked.com\/blog\/nootropics-top-ways-improve-protect-memory\/\"><em>nootropics<\/em><\/a>\u201d community believe that certain compounds and supplements that target this system may have certain \u201ccognitive benefits.\u201d <\/p>\n\n\n\n<p>While the evidence for these compounds having significant effects on cognition in healthy human users is currently relatively weak, this is another common reason that people are sometimes interested in finding out more about substances and compounds that can affect this important neurotransmitter system.<\/p>\n\n\n\n<h3><span id=\"When_to_See_a_Doctor\">When to See a Doctor<\/span><\/h3>\n\n\n\n<p><strong>In any case, if you believe you have a health condition or other reason to try to influence your acetylcholine levels, it is extremely important to always talk to your doctor about any new supplements or dietary changes you make. <\/strong><\/p>\n\n\n\n<p><strong>This is because these approaches could have negative interactions with any other drugs you are taking, other pre-existing health conditions, and other health-related factors. None of the information in this post should ever be used to replace conventional medical treatment.<\/strong><\/p>\n\n\n\n<p><strong>It is also important to keep in mind that many of the compounds and substances discussed below have only been tested in animal- or cell-based studies. This means that their effects and overall safety in healthy human users are not known.<\/strong><\/p>\n\n\n\n<p><strong>Therefore, these compounds should be considered as currently having \u201cinsufficient evidence\u201d for any specific use \u2014 and much more research will be needed to verify what effects they may have in humans, as well as how safe they may be.<\/strong><\/p>\n\n\n\n<h2><span id=\"Factors_That_May_Increase_Acetylcholine\"><strong>Factors That May Increase Acetylcholine<\/strong><\/span><\/h2>\n\n\n\n<p>A large number of different supplements, dietary compounds, and other factors have been proposed to play a role in <a href=\"https:\/\/selfhacked.com\/blog\/cholinergic-activity-natural-boosters\/\">increasing acetylcholine<\/a> levels and activity throughout the brain. For example, some <a href=\"https:\/\/selfhacked.com\/blog\/how-to-increase-serotonin\">serotonin<\/a> neurons are believed to be involved in stimulating the release of acetylcholine. A more comprehensive list of such potential compounds includes:<\/p>\n\n\n\n<ul><li><a href=\"https:\/\/selfhacked.com\/blog\/bacopa-monnieri-4\/\">Bacopa monnieri<\/a> [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/17056129\">R<\/a>]<\/li><li><a href=\"https:\/\/selfhacked.com\/blog\/icariin\/\">Epimedium<\/a> [<a
