From racetams to other experimental compounds, scientists have been wondering if there&#8217;s a pill that can offset cognitive decline and raise our brain&#8217;s potential to another level. And although the research is interesting, the dangers of these compounds are too often downplayed. Read on to uncover the truth. <\/p>\n\n\n<p><!--more--><\/p>\n\n\n<p><strong><em>Disclaimer:<\/em><\/strong><em> This post is <\/em><strong><em>not<\/em><\/strong><em> a recommendation or endorsement for the use of any of the particular compounds or drugs discussed in this post. The FDA has not approved any of these compounds for \u201ccognitive-enhancement\u201d purposes, and the available research on them is still in a very early stage overall. We have written this post for informational purposes only, and our goal is solely to inform people about what science currently says about these substances\u2019 potential uses and mechanisms.<\/em> <\/p>\n\n\n\n<h2><span id=\"Drugs_Proposed_to_Improve_Memory\">Drugs Proposed to Improve Memory<\/span><\/h2>\n\n\n\n<h3><span id=\"INSUFFICIENT_EVIDENCE\">INSUFFICIENT EVIDENCE:<\/span><\/h3>\n\n\n\n<p>Much of the research on the compounds listed below is still in a very early stage, and in most cases, it is not yet possible to come to any firm conclusions about their relative efficacy and safety in human users.<\/p>\n\n\n\n<p>Therefore, the potential effects listed below are still considered to have <strong>insufficient evidence<\/strong>, and these findings should be taken with a grain of salt until further research work &#8211; including large-scale clinical trials in healthy human users &#8211; is performed.<\/p>\n\n\n\n<h3><span id=\"1_Semax\"><strong>1) Semax<\/strong><\/span><\/h3>\n\n\n\n<p><a href=\"https:\/\/selfhacked.com\/blog\/all-about-semax\/\">Semax<\/a> is a drug that has been used in Russia for treating strokes and head injuries, and which has also been claimed to potentially improve learning capacities and memory formation [<a href=\"http:\/\/onlinelibrary.wiley.com\/doi\/10.1111\/j.1471-4159.2006.03658.x\/full\">R<\/a>].<\/p>\n\n\n\n<p><strong>Semax is not approved by the FDA for use in the United States due to a lack of adequate safety and effectiveness data.<\/strong><\/p>\n\n\n\n<p>According to some early studies in animals and humans, some of semax\u2019s reported effects include:<\/p>\n\n\n\n<ul><li>Protecting against <a href=\"https:\/\/selfhacked.com\/blog\/why-you-may-be-suffering-from-low-oxygen-and-not-realize-it\/\">low<\/a> oxygen (<em>hypoxia<\/em>) by promoting the survival of neurons when the brain is not receiving enough oxygen (in rats) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/18756821\">R<\/a>]<\/li><li>Enhancing attention and memory storage [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/18756821\">R<\/a>]<\/li><li>Influencing the formation of newly-learned information and memories [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4757669\/\">R<\/a>]<\/li><li>Increasing selective attention at the moment of receiving information, as well as strengthening and promoting overall learning abilities [<a href=\"http:\/\/onlinelibrary.wiley.com\/doi\/10.1111\/j.1471-4159.2006.03658.x\/full\">R<\/a>]<\/li><li>Improving memory and attention in healthy men under extreme conditions [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/9173745\">R<\/a>]<\/li><\/ul>\n\n\n\n<p>Some of the potential mechanisms that have been suggested to be behind the effects of Semax include:<\/p>\n\n\n\n<ul><li>Increasing <a href=\"https:\/\/selfhacked.com\/blog\/enkephalins\/\"><em>enkephalins<\/em><\/a> (a natural opiate neurotransmitter), which may be involved in memory formation, consolidation, and reactivation\/recall [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/11443939\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/7743204\">R<\/a>]<\/li><li>Enhancing<a href=\"https:\/\/selfhacked.com\/blog\/calcium-101\/\"> calcium<\/a> ion accumulation inside the cells, which may help fight against brain-degenerative processes [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3987924\/\">R<\/a>]<\/li><li>Enhancing the production of key proteins (such as <em>immunoglobulin<\/em>) that are believed to play a role in protecting the brain from stress and damage [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3987924\/\">R<\/a>]<\/li><\/ul>\n\n\n\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Semax is a drug that appeared to improve memory, attention, and learning in Russian studies. However, it was never approved by the FDA due to a lack of proper effectiveness and safety data.<\/div><\/div>\n\n\n\n<h3><span id=\"2_Nicotine\">2<strong>) Nicotine<\/strong><\/span><\/h3>\n\n\n\n<p>Although it is highly addictive and dangerous in most of its common forms,<a href=\"https:\/\/selfhacked.com\/blog\/28-proven-health-benefits-nicotine-4-potential-risks\/\"> nicotine<\/a> is one of the most well-documented drugs to have memory-related effects.<\/p>\n\n\n\n<p><strong>However, we are not recommending starting smoking by any means!<\/strong> Since smoking is a major worldwide cause of death, <strong>the risks of smoking far outweigh any possible benefit.<\/strong> Therefore, we highly advise against smoking or using tobacco &#8211; and if you are already a smoker, seek professional help as soon as possible. <strong>Stopping smoking is among the most important things you can do to improve your health and longevity<\/strong> [<a href=\"https:\/\/www.fda.gov\/tobacco-products\/products-guidance-regulations\/nicotine-addictive-chemical-tobacco-products\">R<\/a>].<\/p>\n\n\n\n<p>While nicotine can also be ingested in other forms &#8211; such as \u201ce-cigarettes\u201d or \u201cvapes\u201d &#8211; the safety of these methods have not been fully proven, and many serious concerns remain about their short-term and long-term effects and safety [<a href=\"https:\/\/www.fda.gov\/tobacco-products\/products-guidance-regulations\/nicotine-addictive-chemical-tobacco-products\">R<\/a>].<\/p>\n\n\n\n<p>Some of the clinical studies on nicotine have relied on other forms, such as nicotine patches or gum. Although these might be \u201csafer\u201d, there is still a high potential for addiction and dependence to nicotine in any form. Therefore, considering how many different &#8211; and relatively much safer &#8211; options there are, it\u2019s probably best to avoid nicotine altogether, and focus instead on less potentially dangerous options (such as the other ones in this post) [<a href=\"https:\/\/www.fda.gov\/tobacco-products\/products-guidance-regulations\/nicotine-addictive-chemical-tobacco-products\">R<\/a>].<\/p>\n\n\n\n<p>With all that in mind, some of the purported cognitive effects of nicotine include:<\/p>\n\n\n\n<ul><li>Makes it easier to consolidate memories in perceptual learning [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/22749926\">R<\/a>]<\/li><li>Improves learning tasks (including contextual conditioning) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2716133\/\">R<\/a>]<\/li><li>May protect the brain [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2716133\/\">R<\/a>]<\/li><li>Improves attention [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3466669\/\">R<\/a>]<\/li><li>Improves dexterity \/ fine motor coordination [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3466669\/\">R<\/a>]<\/li><li>Reduces cognitive impairment [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3466669\/\">R<\/a>]<\/li><li>Improves short-term memory [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/9888618\">R<\/a>]<\/li><\/ul>\n\n\n\n<p>According to one clinical study, nicotine patches reportedly helped alleviate cognitive impairments in Alzheimer\u2019s disease, schizophrenia, and ADD\/ADHD patients [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/11230877\">R<\/a>].<\/p>\n\n\n\n<p>Some other researchers have proposed that nicotine may be a promising treatment for Parkinson\u2019s disease, Down\u2019s syndrome, and age-related memory impairment [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4430096\/\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/16220335\">R<\/a>].<\/p>\n\n\n\n<p>Some of the potential mechanisms that have been suggested to be behind the effects of nicotine include:<\/p>\n\n\n\n<ul><li>Increases acetylcholine activity [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/15370181\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/15652996\">R<\/a>]<\/li><li>Activates the alpha-4 beta-2 nicotinic receptor, which has been implicated in learning [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/11230877\">R<\/a>]<\/li><li>Activates the alpha-7 nicotinic receptor, which has been implicated in long-term memory [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/11230877\">R<\/a>]<\/li><\/ul>\n\n\n\n<p>All in all, while some of the existing research on the potential cognitive effects of nicotine might seem promising, the inherent risk of addiction, dependence, and other significant dangers highly suggest against trying to use nicotine for the purposes of \u201ccognitive enhancement\u201d.<\/p>\n\n\n\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Nicotine might seem like a cognitive enhancer, but it&#8217;s an addictive substance&#8211;the dangers of which far outweigh any potential benefits.<\/div><\/div>\n\n\n\n<h3><span id=\"3_Selegiline\">3<strong>) Selegiline<\/strong><\/span><\/h3>\n\n\n\n<p>In small doses, <a href=\"http:\/\/selegiline\">selegiline<\/a> (<em>L<\/em>&#8211;<em>deprenyl<\/em>) has been reported to inhibit the enzyme <a href=\"https:\/\/selfdecode.com\/gene\/maob\/?utm_source=seo&amp;utm_medium=selfhacked&amp;utm_campaign=id00002\">MAO-B<\/a>. Similarly, in relatively larger doses, some evidence suggests that it may inhibit both MAO-B as well as <a href=\"http:\/\/mao-a\">MAO-A<\/a>.<\/p>\n\n\n\n<p>Because these MAO (<em>monoamine oxidase<\/em>) enzymes are involved in the breakdown of several major types of neurotransmitter in the brain (<em>monoamines<\/em>), inhibiting them can lead to increased levels of several different important neurotransmitters throughout the brain:<\/p>\n\n\n\n<ul><li><strong>MAO-A:<\/strong> <a href=\"https:\/\/selfhacked.com\/blog\/dopamine\/\">dopamine<\/a>, <a href=\"https:\/\/selfhacked.com\/blog\/serotonin-happy-neurotransmitter\/\">serotonin<\/a>, <a href=\"https:\/\/selfhacked.com\/blog\/melatonin\/\">melatonin<\/a>, epinephrine, <a href=\"https:\/\/selfhacked.com\/blog\/norepinephrine-stress-hormone\/\">norepinephrine<\/a>, and tyramine<\/li><li><strong>MAO-B:<\/strong> phenylethylamine, dopamine, and other amines<\/li><\/ul>\n\n\n\n<p>These altered neurotransmitter levels, in turn, could theoretically have a number of effects on the brain and certain cognitive processes.<\/p>\n\n\n\n<p>For example, some early studies in both animals and humans have reported that selegiline may:<\/p>\n\n\n\n<ul><li>Help memory impairments associated with Alzheimer&#8217;s [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC1189014\/\">R<\/a>]<\/li><li>Improve memory and overall cognitive functioning in Parkinson\u2019s patients (vs. placebo) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/10778622\">R<\/a>]<\/li><li>Improve memory impairments via the <em>cholinergic<\/em> system, a major brain system that has been associated with dementia [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/16061218\">R<\/a>]<\/li><li>Improve long-term memory (in aged mice; 0.25 mg\/kg, 3 times per week) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/7997074\">R<\/a>]<\/li><li>Protect against memory impairments from <a href=\"https:\/\/selfhacked.com\/blog\/iron-balance-blood-test-iron-deficiency-anemia-overload\/\">iron<\/a> (in mice) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/labs\/articles\/16122736\/\">R<\/a>]<\/li><\/ul>\n\n\n\n<p>Some of the potential mechanisms that have been suggested to be behind the effects of selegiline include:<\/p>\n\n\n\n<ul><li>Enhancing the activity of the \u201cP300\u201d signal (a brain response associated with vigilance, attention, and decision-making) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/10778622\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/9501758\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/9465283\">R<\/a>]<\/li><li>Increasing dopamine (through inhibiting MAO-B) [<a href=\"https:\/\/pubchem.ncbi.nlm.nih.gov\/compound\/selegiline#section=Pharmacology-and-Biochemistry\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/1946572\">R<\/a>]<\/li><li>Acting as an antioxidant, and protecting against glutamate toxicity (<em>excitotoxicity<\/em>) in neurons [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3320018\/\">R<\/a>]<\/li><li>Inhibiting <a href=\"https:\/\/selfdecode.com\/gene\/ache\/?utm_source=seo&amp;utm_medium=selfhacked&amp;utm_campaign=id00002\"><em>acetylcholinesterase<\/em><\/a> (AChE) &#8211; the enzyme that breaks down the neurotransmitter <a href=\"https:\/\/selfhacked.com\/blog\/acetylcholine\/\">acetylcholine<\/a> &#8211; thereby potentially increasing acetylcholine levels throughout the brain [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3320018\/\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/19941841\">R<\/a>]<\/li><\/ul>\n\n\n\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Selegiline improves memory in some patients with neurodegenerative diseases, but there are no data to suggest it&#8217;s safe or beneficial in healthy people.<\/div><\/div>\n\n\n\n<h3><span id=\"4_Modafinil\">4<strong>) Modafinil<\/strong><\/span><\/h3>\n\n\n\n<p>Modafinil is a medical drug that is FDA-approved for treating certain health conditions, such as narcolepsy and other fatigue-related disorders.<\/p>\n\n\n\n<p><strong>As such, it can only be obtained legally via a doctor\u2019s prescription, and should <em>only<\/em> be used under the ongoing supervision of a qualified medical professional.<\/strong><\/p>\n\n\n\n<p>While this drug is <strong>not<\/strong> approved for any applications related to cognition (or cognitive <em>enhancement<\/em>), some findings from early clinical studies in human patients suggest that this prescription medication may nonetheless have some secondary effects on brain function and cognitive processing.<\/p>\n\n\n\n<p>For example, one study in <a href=\"https:\/\/selfhacked.com\/blog\/depression-genetics-gut-microbes-inflammation\/\">depressed<\/a> patients reported that those who used modafinil showed better episodic and working memory [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/28299368\">R<\/a>].<\/p>\n\n\n\n<p>According to another study, adult ADHD patients who were administered<a href=\"https:\/\/selfhacked.com\/blog\/modafinil\/\"> modafinil<\/a> (200 mg\/day) were reported to show significant improvements in short-term memory, visual memory, and spatial planning. However, although they responded more accurately on tests, they had slower responses overall &#8211; which may suggest that there are some important trade-offs involved in some of modafinil\u2019s potential cognitive effects [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/15121488\">R<\/a>].<\/p>\n\n\n\n<p>Similarly, modafinil has been reported to improve working memory in individuals with cognitive impairments related to chronic methamphetamine abuse [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/20653641\">R<\/a>].<\/p>\n\n\n\n<p>While some of these early results seem somewhat promising, it is important to note that all of these studies involved human patients with very specific medical conditions or disorders, and so there is no direct evidence as yet that this drug would have similar effects in healthy human users.<\/p>\n\n\n\n<p>Some of the potential mechanisms that have been suggested to be behind the effects of modafinil include:<\/p>\n\n\n\n<ul><li>Increases glutamate in the thalamus [<a href=\"http:\/\/ucdirc.ucdavis.edu\/people\/papers\/minzenberg_carter_N2007.pdf\">R<\/a>]<\/li><li>Prevents glutamate toxicity (<em>excitotoxicity<\/em>), which damages and kills neurons [<a href=\"http:\/\/ucdirc.ucdavis.edu\/people\/papers\/minzenberg_carter_N2007.pdf\">R<\/a>]<\/li><li>Increases dopamine, norepinephrine (inhibits dopamine\/norepinephrine transporter) and serotonin [<a href=\"http:\/\/ucdirc.ucdavis.edu\/people\/papers\/minzenberg_carter_N2007.pdf\">R<\/a>]<\/li><li>Increases <a href=\"http:\/\/selfhacked.com\/blog\/histamine\/\">histamine<\/a> neurons in multiple parts of the brain [<a href=\"http:\/\/ucdirc.ucdavis.edu\/people\/papers\/minzenberg_carter_N2007.pdf\">R<\/a>]<\/li><\/ul>\n\n\n\n<p>However, while it is possible to find and order this drug online, it is not advised to experiment with it casually, as it may not be legal to possess where you live. It also has some serious potential side-effects and other risks, which is why it is typically only available by a doctor\u2019s prescription. Taking all of this into account, if you are interested in using supplements to potentially enhance your cognitive abilities, you are better off sticking to many of the safer and legal options discussed elsewhere in this post!<\/p>\n\n\n\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Modafinil is FDA-approved for narcolepsy&#8211;the only indication this drug has been properly researched for. It is also one of the most \u201cpopular\u201d nootropic compounds, and as such there have been many claims made about its potential effects. But what does the current science really say?<\/p>\n\n\n\n<p>So far, the available data and hard evidence from legitimate clinical studies of this drug are a bit modest &#8211; at least compared to a lot of the hype that you can find about piracetam online. This isn\u2019t to say that there isn\u2019t any evidence, though; just that the full picture is still complex, with many mixed and inconclusive results so far.<\/p>\n\n\n\n<p>We\u2019ll briefly discuss some of the more promising findings behind it below: but if you want to get a more complete picture of the current science behind this drug &#8211; as well as some of its potential side-effects and other risks &#8211; we would encourage you to check out our other, more detailed SelfHacked posts on it <a href=\"https:\/\/selfhacked.com\/blog\/top-12-benefits-piracetam\/\">here<\/a> and <a href=\"https:\/\/selfhacked.com\/blog\/why-i-dont-supplement-with-piracetam\/\">here<\/a>.<\/p>\n\n\n\n<p>According to one early human study, piracetam was reported to improve long-term\/short-term memory retrieval in 60 dyslexic boys [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/3522510\">R<\/a>].<\/p>\n\n\n\n<p>Piracetam was also associated with a significant improvement in verbal and nonverbal short-term memory and attention in patients undergoing bypass surgery [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/12806565\">R<\/a>]. However, this was a small study, and only dealt with patients with a specific underlying medical condition &#8211; so we can\u2019t assume that similar effects would necessarily apply to healthy human users without appropriate follow-up studies to confirm this.<\/p>\n\n\n\n<p>Additionally, a couple of early animal studies have reported that chronic treatment (250 mg\/kg) with piracetam significantly improved working memory in mice with drug-induced amnesia. However, in one of these studies, piracetam was combined with <a href=\"https:\/\/selfhacked.com\/blog\/citicoline\/\">citicoline<\/a>, so it\u2019s not possible to determine which specific compound was responsible for these effects [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/9533177\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/2392950\">R<\/a>].<\/p>\n\n\n\n<p>Some of the potential mechanisms that have been suggested to be behind the effects of piracetam include:<\/p>\n\n\n\n<ul><li>Increasing the function of muscarinic choline receptors [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/16459490\">R<\/a>]<\/li><li>Stimulating NMDA receptors [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/16007238\">R<\/a>]<\/li><li>Improving cell membrane permeability [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/16007238\">R<\/a>]<\/li><li>Enhancing the brain\u2019s ability to use<a href=\"https:\/\/selfhacked.com\/blog\/glucose\/?utm_source=seo&amp;utm_medium=selfhacked&amp;utm_campaign=id00002\"> glucose<\/a><\/li><\/ul>\n\n\n\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">No valid evidence supports the use of piracetam as a nootropic. The results so far have been inconclusive.<\/div><\/div>\n\n\n\n<h3><span id=\"6_Phenylpiracetam_Phenotropil\">6<strong>) Phenylpiracetam <\/strong>(\u201c<strong>Phenotropil<\/strong>\u201d)<\/span><\/h3>\n\n\n\n<p><a href=\"https:\/\/selfhacked.com\/blog\/phenylpiracetam-dosage\/\"><em>Phenylpiracetam<\/em><\/a> is a Russian drug developed in 1983 to combat the prolonged stress of working in space. Today it is a prescription medication typically sold under the name \u201c<em>Phenotropil<\/em>\u201d in Russia.<\/p>\n\n\n\n<p>It is not approved elsewhere (such as by the FDA in the USA) for any specific use.<\/p>\n\n\n\n<p>According to one preliminary study, phenotropil was reported to improve daily functioning and brain function in stroke patients [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/21626817\">R<\/a>].<\/p>\n\n\n\n<p>In another study on patients with brain damage (<em>encephalopathy<\/em>), phenotropil was reported to improve brain functions, memory, and counting ability [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/16447562\">R<\/a>].<\/p>\n\n\n\n<p>In animals that lack blood flow to the brain, phenotropil was reported to improve their memory functions and vitality [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/17523446\">R<\/a>].<\/p>\n\n\n\n<p>However, it is important to notice that <em>all<\/em> of the above studies were done in subjects with various underlying forms of brain damage, and so it cannot be concluded that phenotropil would have similar effects in healthy users on the basis of any of these findings.<\/p>\n\n\n\n<p>The mechanisms of phenylpiracetam\/phenotropil are not clinically clear but should &#8211; in theory &#8211; be similar to other drugs in the <a href=\"https:\/\/selfhacked.com\/blog\/comparison-aniracetam-oxiracetam-phenylracetam-pramiracetam\/\"><em>racetam<\/em> family<\/a>.<\/p>\n\n\n\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Phenylpiracetam was allegedly invented by Russian scientists to combat the stress of working in space. The FDA hasn&#8217;t approved it due to a lack of proper human trials.<\/div><\/div>\n\n\n\n<h3><span id=\"7_Oxiracetam\">7<strong>) Oxiracetam<\/strong><\/span><\/h3>\n\n\n\n<p><em>Oxiracetam<\/em> is another <a href=\"https:\/\/selfhacked.com\/blog\/comparison-aniracetam-oxiracetam-phenylracetam-pramiracetam\/\"><em>racetam<\/em><\/a> drug with some early results showing potential effects on cognitive function.<\/p>\n\n\n\n<p>For example, in a study of patients with Alzheimer\u2019s or multi-infarct dementia, 800 mg <a href=\"https:\/\/selfhacked.com\/blog\/oxiracetam\/\">oxiracetam<\/a> twice daily was reported to improve memory, with no signs of tolerance developed. However, performances on some cognitive tests were statistically <em>worse<\/em> than baseline after late-stage follow-up, indicating very mixed results [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/1603291\">R<\/a>].<\/p>\n\n\n\n<p>One study in humans reported that oxiracetam\u2019s effects on memory may be greater compared to piracetam [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/3594458\">R<\/a>].<\/p>\n\n\n\n<p>In a study on rats and mice, oxiracetam was reported to improved memory as well as piracetam. In addition, it also reported improved these animals\u2019 learning abilities [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/3594453\">R<\/a>].<\/p>\n\n\n\n<p>According to one rat study, oxiracetam reportedly reduced brain injury, and increased learning, memory, and spatial cognition in rats with traumatic brain injuries [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24175546\">R<\/a>].<\/p>\n\n\n\n<p>Some of the potential mechanisms that have been suggested to be behind the effects of oxiracetam include:<\/p>\n\n\n\n<ul><li>Prevents decreases in the brain\u2019s levels of <a href=\"https:\/\/selfhacked.com\/blog\/acetylcholine\/\">acetylcholine<\/a>, a neurotransmitter that is believed to be critical in overall memory function [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/3659072\">R<\/a>].<\/li><\/ul>\n\n\n\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Oxiracetam was researched in a small group of Alzheimer&#8217;s patients, but its effects are still uncertain.<\/div><\/div>\n\n\n\n<h3><span id=\"8_Pramiracetam\">8<strong>) Pramiracetam<\/strong><\/span><\/h3>\n\n\n\n<p><a href=\"https:\/\/selfhacked.com\/blog\/comparison-aniracetam-oxiracetam-phenylracetam-pramiracetam\/\"><em>Pramiracetam<\/em><\/a> is marketed as a potential treatment for memory and attention deficits in aging people with neurodegenerative diseases and vascular dementia. However, its evidence for efficacy in healthy human subjects remains somewhat limited.<\/p>\n\n\n\n<p>In one study of males with brain injuries, pramiracetam (400 mg) was reported to improve memory (with no build-up of tolerance) over an 18-month period. The effect was reported to be consistent up to 1 month after discontinuing treatment [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/1786500\">R<\/a>].<\/p>\n\n\n\n<p>Pramiracetam has also been reported to partially counteract experimentally-induced amnesia (caused by scopolamine) in one study of both young and old (18-42 and 55-65-year-old) human participants [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/15374306\">R<\/a>].<\/p>\n\n\n\n<p>According to one other human study, pramiracetam was reported to have a statistically significant effect on improving the memory of patients suffering from lack of oxygen in the brain (<em>chronic cerebral blood insufficiency<\/em>) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/m\/pubmed\/14965012\/?i=6&amp;from=Pramiracetam\">R<\/a>].<\/p>\n\n\n\n<p>However, all of these human studies were done on people with brain injuries or experimentally-induced cognitive deficits, and so cannot be translated over into healthy human users without much additional research.<\/p>\n\n\n\n<p>In one animal study on rats, pramiracetam (7.5 mg\/kg and 15 mg\/kg) was reported to improve long-term memory, but did not significantly improve working memory [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/3088666\">R<\/a>].<\/p>\n\n\n\n<p>Some of the potential mechanisms that have been suggested to be behind the effects of pramiracetam include:<\/p>\n\n\n\n<ul><li>Increases nicotinic <a href=\"https:\/\/selfhacked.com\/blog\/acetylcholine\/\">acetylcholine<\/a> receptor activity [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/20166767\">R<\/a>].<\/li><li>Increases <a href=\"https:\/\/selfhacked.com\/blog\/top-18-scientific-health-benefits-choline-cdp-choline-alpha-gpc\/\">choline<\/a> uptake (in rat hippocampus) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/20166767\">R<\/a>].<\/li><\/ul>\n\n\n\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Pramiracetam seems to improve memory in people with cognitive decline, but its purported nootropic effects in healthy people remain unproven.<\/div><\/div>\n\n\n\n<h3><span id=\"9_PRL-8-53\">9) PRL-8-53<\/span><\/h3>\n\n\n\n<p><a href=\"https:\/\/selfhacked.com\/blog\/prolactin\/\">PRL<\/a>-8-53 is an experimental nootropic research drug. As such it is extremely new, and very little is known about what its potential effects or mechanisms might be.<\/p>\n\n\n\n<p>In one double-blind study, PRL-8-53 was reported to enhance learning and memory retention in humans (at very low doses) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/418433\">R<\/a>].<\/p>\n\n\n\n<p>The Mechanisms of PRL-8-53 have not been studied yet, and currently, remain unclear.<\/p>\n\n\n\n<h3><span id=\"Animal_And_Cell_Research_LACKING_EVIDENCE\">Animal And Cell Research (LACKING EVIDENCE):<\/span><\/h3>\n\n\n\n<p>For the compounds listed below, what we currently know about their potential effects is based <em>solely<\/em> on animal- or cell-based studies, and are <strong>lacking evidence<\/strong> from any appropriate human trials so far. Therefore, these are only potential \u201claunching-points\u201d for future clinical studies in humans, and no solid conclusions can be made about these effects in human users until much more additional research is done.<\/p>\n\n\n\n<h3><span id=\"10_Fasoracetam_NS-105\">10) Fasoracetam (\u201cNS-105\u201d)<\/span><\/h3>\n\n\n\n<p><a href=\"https:\/\/selfhacked.com\/blog\/fasoracetam\/\"><em>Fasoracetam<\/em><\/a> &#8211; also sometimes referred to as \u201cNS-105\u201d &#8211; is another member of the <a href=\"https:\/\/selfhacked.com\/blog\/comparison-aniracetam-oxiracetam-phenylracetam-pramiracetam\/\"><em>racetam<\/em> family<\/a> of drugs. It is currently undergoing clinical trials for the treatment of vascular dementia and ADHD, although it is most likely still a long ways off from possibly becoming FDA-approved for any particular official use.<\/p>\n\n\n\n<p>The mechanisms of fasoracetam are not clinically clear, but are theoretically similar to those of other drugs in the racetam family, due to the chemical and molecular similarities they appear to share.<\/p>\n\n\n\n<p>Apart from one preliminary animal study (below), large-scale clinical studies on this drug are generally lacking, and so not much is known for sure about how effective or safe it may be in healthy human users &#8211; especially over the long term.<\/p>\n\n\n\n<p>According to one early animal study, fasoracetam was reported to partially reverse memory impairments caused by <a href=\"https:\/\/selfhacked.com\/blog\/baclofen\/\"><em>baclofen<\/em><\/a> (a potent <a href=\"https:\/\/selfhacked.com\/blog\/natural-ways-to-increase-gaba\/\">GABA<\/a> B agonist drug) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/10494996\">R<\/a>].<\/p>\n\n\n\n<p>However, this is only one study, in rats, and so a lot more research will clearly be needed to understand more about this drug\u2019s potential effects in humans. As such, it\u2019s not recommended to experiment with this drug, since its efficacy and safety remain unknown.<\/p>\n\n\n\n<p>&gt;&gt;&gt; <em>To learn more about fasoracetam, check out our detailed SelfHacked post on it <\/em><a href=\"https:\/\/selfhacked.com\/blog\/fasoracetam\/\"><em>here<\/em><\/a><em>.<\/em><\/p>\n\n\n\n<h3><span id=\"11_Coluracetam_BCI-540_Mkc-231\">11) Coluracetam (\u201cBCI-540\u201d \/ \u201cMkc-231\u201d)<\/span><\/h3>\n\n\n\n<p><em>Coluracetam<\/em> is yet another drug variant in the <a href=\"https:\/\/selfhacked.com\/blog\/comparison-aniracetam-oxiracetam-phenylracetam-pramiracetam\/\"><em>racetam<\/em><\/a> family with a couple of animal studies on its potential effects. However, no human studies have been performed yet.<\/p>\n\n\n\n<p>In one animal study in rats with brain damage from chemicals, coluracetam was reported to reduce the toxically-induced memory impairments, with no apparent adverse side-effects [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/8740080\">R<\/a>].<\/p>\n\n\n\n<p>In one other animal study, coluracetam was also reported to induce long-lasting (72-hour) effects on cognitive function after repeated administration (rat study) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/18461272\">R<\/a>].<\/p>\n\n\n\n<p>Some of the potential mechanisms that have been suggested to be behind the effects of coluracetam include:<\/p>\n\n\n\n<ul><li>Increasing <a href=\"https:\/\/selfhacked.com\/blog\/top-18-scientific-health-benefits-choline-cdp-choline-alpha-gpc\/\">choline<\/a> uptake [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/8740080\">R<\/a>].<\/li><li>Reversing hippocampal <a href=\"https:\/\/selfhacked.com\/blog\/acetylcholine\/\">acetylcholine<\/a> depletion [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/7710736\">R<\/a>].<\/li><\/ul>\n\n\n\n<h3><span id=\"12_Selank\">12) Selank<\/span><\/h3>\n\n\n\n<p><a href=\"https:\/\/selfhacked.com\/blog\/selank-effects\/\">Selank<\/a> is another compound that was originally developed in Russia, and which has been reported to be a <em>nootropic<\/em> and <a href=\"https:\/\/selfhacked.com\/blog\/natural-treatments-anxiety\/\">anxiety<\/a>-reducing peptide. It is a synthetic analog of a component of <em>immunoglobulin G<\/em> (<em>tuftsin<\/em>).<\/p>\n\n\n\n<p>However, the evidence in support of its effects, while somewhat promising, has only been shown in animals so far.<\/p>\n\n\n\n<p>For example, according to some early findings, selank reportedly improved memory and brain function in rats with lowered learning abilities. The studied rats were also reported to have enhanced memory abilities under conditions of high emotional stress [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/12449836\">R<\/a>].<\/p>\n\n\n\n<p>In another study in rats, injecting 300 \u03bcg\/kg of selank was reported to increase the stability of memory traces (newly-formed memories) for up to one month [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/20919548\">R<\/a>].<\/p>\n\n\n\n<p>Finally, selank (300 \u03bcg\/kg) was reported to help restore certain cognitive functions (such as memory, learning, and attention to sensory stimuli) in rats with chronic artificial inhibition of the brain (specifically, the cerebral catecholaminergic system) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/18683497\">R<\/a>].<\/p>\n\n\n\n<p>Some of the potential mechanisms that have been suggested to be behind the effects of Selank include:<\/p>\n\n\n\n<ul><li>Activates <a href=\"https:\/\/selfhacked.com\/blog\/dopamine\/\">the dopamine<\/a> D5 receptor (<a href=\"https:\/\/selfdecode.com\/gene\/drd5\/?utm_source=seo&amp;utm_medium=selfhacked&amp;utm_campaign=id00002\">DRD5<\/a>), which plays a key role in the formation of memory and learning processes [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4757669\/\">R<\/a>].<\/li><li>Increases <a href=\"https:\/\/selfhacked.com\/blog\/a-comprehensive-list-of-natural-ways-to-increase-bdnf\/\">BDNF<\/a> in the hippocampus of rats [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/18841804\">R<\/a>].<\/li><li>Increases <em>enkephalins<\/em>, which play a role in memory formation, consolidation, and reactivation\/recall [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/11443939\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/7743204\">R<\/a>].<\/li><\/ul>\n\n\n\n<p>However, all of these preliminary findings will have to be followed up on by large-scale human studies in order to fully confirm these potential effects.<\/p>\n\n\n\n<div class=\"sh-summary\"><div class=\"sh-summary-content\"><\/p>\n\n\n\n<h3><span id=\"Takeaway\">Takeaway<\/span><\/h3>\n\n\n\n<p>Only a handful of drugs have been properly researched for their alleged nootropic effects, while all others lack scientific ground. <\/p>\n\n\n\n<p>On top of that, the existing research suffers major flaws. For example, some drugs that are claimed to be nootropics in Russia (like Semax and Phenylpiracetam) never underwent proper trials in healthy humans. For this reason, they have not been approved by the FDA.<\/p>\n\n\n\n<p>Other purported nootropics are downright dangerous, such as nicotine.
