This system has three groups or classes;<a href=\"https:\/\/selfdecode.com\/gene\/hla-b\/\"> HLA-B<\/a> genes, along with HLA-A and HLA-C, belong to the class I [<a href=\"https:\/\/ghr.nlm.nih.gov\/gene\/HLA-B\">R<\/a>].<\/p>\n<p>HLAs are proteins or antigens on the surface of<a href=\"https:\/\/selfhacked.com\/blog\/how-to-increase-and-decrease-white-blood-cells\/\"> white blood cells<\/a>. They help flag and remove external components that may cause infection. HLA-B antigens bring bacterial or viral proteins from the inside to the cell surface so T-lymphocytes can detect and kill the infected cell [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC1079257\/\">R<\/a>].<\/p>\n<p>They also have anti-cancer effects and play a role in organ transplant rejection [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4333229\/\">R<\/a>,<a href=\"https:\/\/ghr.nlm.nih.gov\/gene\/HLA-B\"> R<\/a>].<\/p>\n<p>HLA genes come in many different forms or variants, which enable the fine-tuning of your immune system. Their diversity can be a double-edged sword, however, as some types increase the risk of autoimmune disorders [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2628004\/\">R<\/a>].<\/p>\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">The HLA genes encode proteins (antigens) that bring external components to the surface of white blood cells, enabling your immune system to detect and destroy them.<\/div><\/div>\n<h3><span id=\"HLA-B27_Antigen\"><a id=\"post-80994-_692bfyft1ss9\"><\/a><strong>HLA-B27 Antigen<\/strong><\/span><\/h3>\n<p>The HLA-B27 antigen supports antiviral immune response by \u201cflagging\u201d peptides from viruses &#8211; such as influenza, HIV, and Epstein-Barr &#8211; and presenting them to T-killer cells [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3240147\/\">R<\/a>].<\/p>\n<p>The gene for HLA-B27 has over 100 variations. Despite the beneficial roles, HLA-B27 is strongly associated with a group of autoimmune joint disorders known as <em>spondyloarthritis<\/em> (SpA) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/gene\/3106\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC5364987\/\"> R<\/a>].<\/p>\n<p>The most common variations associated with autoimmunity are [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/23990399\">R<\/a>]:<\/p>\n<ul>\n<li>HLA-B*27:05 (Caucasians\/whites)<\/li>\n<li>HLA-B*27:04 (Chinese)<\/li>\n<li>HLA-B*27:02 (Mediterranean)<\/li>\n<\/ul>\n<p>On the other hand, HLA-B*27:06 seems to have a protective role [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/22324941\">R<\/a>].<\/p>\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">HLA-B27 enables an antiviral immune response, but certain subtypes correlate with autoimmune disorders.<\/div><\/div>\n<h4><a id=\"post-80994-_xpi37p5qz0ao\"><\/a><strong>Prevalence<\/strong><\/h4>\n<p>The prevalence of HLA-B27 varies from 50% in some Indian populations to almost zero in Australian Aborigines. About <strong>6% of the US population<\/strong> carries this variant, which is more common among white people [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4038331\/\">R<\/a>].<\/p>\n<h2><span id=\"HLA-B27_and_Autoimmune_Diseases\"><a id=\"post-80994-_ah8echamwf30\"><\/a><strong>HLA-B27 and Autoimmune Diseases<\/strong><\/span><\/h2>\n<p><em>It\u2019s important to note that just because certain genotypes are associated with a disease, it doesn\u2019t necessarily mean that everyone with that genotype will actually develop the disease! Many different factors, including other genetic and environmental factors, can influence the risk of autoimmune diseases.<\/em><\/p>\n<h3><span id=\"Ankylosing_Spondylitis_AS\"><a id=\"post-80994-_6mra55t6jyyr\"><\/a><strong>Ankylosing Spondylitis (AS)<\/strong><\/span><\/h3>\n<p>As mentioned, HLA-B27 strongly correlates with higher rates of certain autoimmune diseases, such as [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/4604133\/\">R<\/a>]:<\/p>\n<ul>\n<li>Psoriasis<\/li>\n<li>Ankylosing spondylitis (spine deformation)<\/li>\n<li>IBD (inflammatory bowel disease), in combination with spondylitis<\/li>\n<li>Reactive arthritis (Reiter\u2019s syndrome) &#8211; inflammation of joints, urethra, and eyes<\/li>\n<\/ul>\n<p>These conditions have a common name <em>spondyloarthritis<\/em>, the main one being <strong>ankylosing spondylitis<\/strong> (AS). In AS, inflammation gradually fuses spine vertebrae, causing back<a href=\"https:\/\/selfhacked.com\/blog\/23-ways-to-combat-pain-naturally-by-increasing-your-opioids\/\"> pain<\/a> and limited movement. It usually affects young men [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/25447326\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/books\/NBK470173\/\"> R<\/a>].<\/p>\n<p>Doctors treat AS with drugs such as NSAIDs and TNF-inhibitors, but their effectiveness is limited. Besides HLA-B27, other genetic and environmental factors are involved [<a href=\"https:\/\/onlinelibrary.wiley.com\/doi\/full\/10.1002\/art.39298\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3335985\/\"> R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC1079257\/\"> R<\/a>].<\/p>\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">HLA-B27 is associated with autoimmune disorders known as spondyloarthritis. The main one is ankylosing spondylitis, which causes spine inflammation and chronic back pain.<\/div><\/div>\n<h3><span id=\"The_Connection\"><a id=\"post-80994-_eporzqclf5np\"><\/a><strong>The Connection<\/strong><\/span><\/h3>\n<p>The connection between HLA-B27 and ankylosing spondylitis (AS) is among the strongest in the HLA system: <strong>90\u201395%<\/strong> of all AS patients have this variant. For other conditions, the prevalence ranges between 50-90% [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC1079257\/\">R<\/a>].<\/p>\n<p>Does this mean almost everyone with HLA-B27 will get AS or another autoimmune disease?
