It is produced by the body\u2019s fat cells [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/12439643\">R<\/a>].<\/p>\n<p>When a person eats enough food, leptin is released into the bloodstream, where it travels to the brain, signaling that they are full and that can burn calories at a normal rate [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/10766253\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/25232147\"> R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4454393\/\"> R<\/a>].<\/p>\n<p>To learn more about normal leptin levels and what this hormone does in the body,<a href=\"https:\/\/selfhacked.com\/blog\/all-about-leptin%E2%80%A6n-cfs-and-weight\/\"> read the first part of this series<\/a>.<\/p>\n<p>This post will focus on the dangers of high leptin. Elevated leptin levels are associated with obesity, overeating, and inflammation-related diseases, including high blood pressure, metabolic syndrome, and heart disease [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/21957899\">R<\/a>].<\/p>\n<h3><span id=\"Leptins_Role_in_Weight\"><a id=\"post-77608-_xbfpw1tu39rp\"><\/a><strong>Leptin\u2019s Role in Weight<\/strong><\/span><\/h3>\n<p>To recap, leptin is one of the big 4 hormones that determine weight. It is made in fat tissue, while receptors for it are dense in two important brain regions (the <a href=\"https:\/\/selfhacked.com\/blog\/hypothalamus-101\/\">hypothalamus<\/a> and hippocampus) [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2805706\/\">R<\/a>].<\/p>\n<p>Leptin acts as a signal to the brain to inhibit food intake and enable the storage of surplus calories (in fat cells).<\/p>\n<p>At the same time, it protects non-fatty tissue from fat overload. Increased leptin is associated with higher body fat mass, a larger size of individual fat cells, overeating, and excessive <a href=\"https:\/\/selfhacked.com\/blog\/top-10-ways-to-curb-your-hunger-and-approaches-that-didnt-work\/\">hunger<\/a>. In rodents, it increases energy expenditure by using brown fat for energy [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3409346\/#R30\">R<\/a>].<\/p>\n<p>The more fat is present, the more leptin is produced. Autoimmune diseases are associated with increased secretion of leptin [<a href=\"http:\/\/www.hindawi.com\/journals\/mi\/2010\/568343\/\">R<\/a>]. In terms of both structure and function, leptin resembles <a href=\"http:\/\/selfhacked.com\/blog\/interleukin-6\/\">IL-6<\/a>.<\/p>\n<p>Leptin increases the release of proinflammatory cytokines <a href=\"http:\/\/selfhacked.com\/blog\/supplements-lifestyle-factors-influence-tnf-interleukin-6-il-6\/\">TNF-alpha<\/a>, <a href=\"http:\/\/selfdecode.com\/gene\/il2\/?utm_source=seo&#038;utm_medium=selfhacked&#038;utm_campaign=id00002\">IL-2<\/a>,<a href=\"https:\/\/selfhacked.com\/blog\/interleukin-6\/\"> IL-6<\/a>, and <a href=\"http:\/\/selfhacked.com\/blog\/interleukin-1\/\">IL-1<\/a><a href=\"https:\/\/selfdecode.com\/gene\/IL12a\/?utm_source=seo&#038;utm_medium=selfhacked&#038;utm_campaign=id00002\">2<\/a> [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3637529\/#B27\">R<\/a>].<\/p>\n<p>It also increases<a href=\"https:\/\/selfhacked.com\/blog\/c-reactive-protein\/\"> C-Reactive Protein<\/a> (<a href=\"https:\/\/selfhacked.com\/blog\/c-reactive-protein\/\">CRP<\/a>) [<a href=\"http:\/\/www.hindawi.com\/journals\/isrn\/2013\/139239\/#B54\">R<\/a>].<\/p>\n<p>Leptin provokes the release of proinflammatory cytokines from many cell types, including microglia in the brain [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3637529\/#B12\">R<\/a>], and is a mediator of cytokine-induced sickness behavior (CFS) [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3637529\/#B13\">R<\/a>].<\/p>\n<p>Multiple studies have demonstrated elevated levels of leptin in chronic inflammatory conditions [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3637529\/#B29\">R<\/a>].<\/p>\n<p>It also increases both red and<a href=\"https:\/\/selfhacked.com\/blog\/how-to-increase-and-decrease-white-blood-cells\/\"> white blood cells<\/a> [<a href=\"http:\/\/www.hindawi.com\/journals\/mi\/2010\/568343\/\">R<\/a>].<\/p>\n<p>Elevated leptin is associated with higher <a href=\"https:\/\/selfhacked.com\/blog\/how-to-increase-and-decrease-white-blood-cells\/\">white blood cell<\/a> (<a href=\"https:\/\/selfhacked.com\/blog\/how-to-increase-and-decrease-white-blood-cells\/\">WBC<\/a>) counts in both men and women [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/15724240\">R<\/a>].<\/p>\n<h4><a id=\"post-77608-_7g8yjpz2t835\"><\/a><strong>2) Fatigue<\/strong><\/h4>\n<p>Leptin is believed to cause inflammatory fatigue [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3637529\/#B10\">R<\/a>]. Leptin is higher in women even when controlling for weight, and women are more likely to be diagnosed with chronic fatigue syndrome (CFS) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/11244464\">R<\/a>].<\/p>\n<p>This is believed to be, in part, because<a href=\"https:\/\/selfhacked.com\/blog\/testosterone-will-add\/\"> testosterone<\/a> suppresses leptin in men (clinical trials) [<a href=\"http:\/\/press.endocrine.org\/doi\/full\/10.1210\/jcem.83.9.5134\">R<\/a>].<\/p>\n<p>Leptin increases were associated with greater fatigue scores in people with CFS. Some researchers believe that it plays a causal role in CFS; however, chronic fatigue is a controversial and poorly understood condition, and this conclusion is by no means unanimous [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/23570606\">R<\/a>].<\/p>\n<p>Leptin levels in the blood are also associated with fatigue severity in patients with chronic hepatitis C and<a href=\"https:\/\/selfhacked.com\/blog\/causes-ibs-general-gut-dysfunction\/\"> irritable bowel syndrome<\/a> (<a href=\"https:\/\/selfhacked.com\/blog\/causes-ibs-general-gut-dysfunction\/\">IBS<\/a>) [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3637529\/#B10\">R<\/a>].<\/p>\n<p>In people diagnosed with CFS, leptin increases more in response to <a href=\"https:\/\/selfhacked.com\/blog\/need-know-cortisol-health-effects\/\">cortisol<\/a> [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/11453960\">R<\/a>].<\/p>\n<h4><a id=\"post-77608-_32c1l4e71v33\"><\/a><strong>3) Th1 Immunity<\/strong><\/h4>\n<p>Leptin activates the <a href=\"https:\/\/selfdecode.com\/gene\/jak2\/?utm_source=seo&#038;utm_medium=selfhacked&#038;utm_campaign=id00002\">JAK<\/a><a href=\"https:\/\/selfdecode.com\/gene\/jak2\/?utm_source=seo&amp;utm_medium=selfhacked&amp;utm_campaign=id00002\">2<\/a>\u2013 <a href=\"http:\/\/selfhacked.com\/blog\/natural-stat3-inhibitors\/\">STAT3<\/a> pathway [<a href=\"http:\/\/press.endocrine.org\/doi\/10.1210\/jcem.86.2.7245\">R<\/a>] and <strong>promotes<\/strong><a href=\"https:\/\/selfhacked.com\/blog\/supplements-foods-exercise-right-type-th1-vs-th2-dominance\/\"><strong> Th1 responses<\/strong><\/a> [<a href=\"http:\/\/www.hindawi.com\/journals\/mi\/2010\/568343\/\">R<\/a>].<\/p>\n<p>By stimulating STAT3, it might also stimulate <a href=\"http:\/\/selfhacked.com\/blog\/th17\/\">Th17<\/a><a href=\"https:\/\/selfhacked.com\/blog\/th17\/\"> responses<\/a> [<a href=\"http:\/\/www.jimmunol.org\/content\/179\/7\/4313.full.pdf\">R<\/a>]. Leptin also changed human dendritic cells (DCs), directing them towards Th1 dominance [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/17125143?dopt=AbstractPlus\">R<\/a>].<\/p>\n<p>It was also found to be necessary for Th1 dependent inflammatory processes, acting as a critical regulator of CD4 T cells [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/19059800?dopt=AbstractPlus\">R<\/a>].<\/p>\n<p>The effect of leptin polarizing T cells towards a Th1 response seems to be mediated by stimulating the synthesis of<a href=\"http:\/\/selfdecode.com\/gene\/il2\/?utm_source=seo&amp;utm_medium=selfhacked&amp;utm_campaign=id00002\"> IL-2<\/a>, IL-12, and <a href=\"https:\/\/selfhacked.com\/blog\/interferon-gamma-how-to-increase-decrease-high-low-levels\/\">IFN-gamma<\/a> (all Th1 cytokines) and inhibiting the production of <a href=\"http:\/\/selfhacked.com\/blog\/il-10\/\">IL-10<\/a> and <a href=\"https:\/\/selfdecode.com\/gene\/il4\/?utm_source=seo&#038;utm_medium=selfhacked&#038;utm_campaign=id00002\">IL-4<\/a> [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/15905523\">R<\/a>,<a href=\"https:\/\/doi.org\/10.1046%2fj.1365-2249.2002.01900.x\"> R<\/a>].<\/p>\n<p>Th1 dominance correlates with excess leptin in this study of dialysis patients [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/19966187?dopt=AbstractPlus\">R<\/a>]. If you\u2019re <a href=\"http:\/\/selfhacked.com\/blog\/supplements-people-th2-dominant\/\">Th2<\/a> dominant, it doesn\u2019t mean leptin isn\u2019t causing problems. It simply means that it\u2019s causing <em>different<\/em> problems.<\/p>\n<h4><a id=\"post-77608-_sakfjeu72swf\"><\/a><strong>4) Mast Cells<\/strong><\/h4>\n<p>Mast cells in the human skin, lungs, gut, and the urogenital tract contain leptin and leptin receptors [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/19241089?dopt=AbstractPlus\">R<\/a>], suggesting that leptin communicates with mast cells.<\/p>\n<p>It seems like leptin causes mast cells to be more inflammatory [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/26481668\">R<\/a>].<\/p>\n<p>In metabolic syndrome patients, there was a positive correlation between leptin levels and the number of fat tissue mast cells, suggesting that leptin may stimulate mast cells [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/11935372\">R<\/a>].<\/p>\n<p>Leptin correlates with (probably increases) mast cell activation in children with asthma who exercised [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4406702\/\">R<\/a>].<\/p>\n<p>It suppresses appetite, in part, by increasing<a href=\"https:\/\/selfhacked.com\/blog\/histamine\/\"> histamine<\/a> release and activating histamine H1 receptors in the hypothalamus [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/11272151\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/10580415\"> R<\/a>].<\/p>\n<h4><a id=\"post-77608-_dw597k8gmt7d\"><\/a><strong>5) Regulatory T cells<\/strong><\/h4>\n<p>Leptin impedes the production of <a href=\"https:\/\/selfhacked.com\/blog\/treg\/\">Treg cells<\/a>. Tregs are a critical part of the immune system that creates a tolerance to agents that would otherwise trigger an immune response [<a href=\"http:\/\/www.hindawi.com\/journals\/mi\/2010\/568343\/\">R<\/a>].<\/p>\n<h4><a id=\"post-77608-_jkabub2yv1t\"><\/a><strong>6) HPA Axis<\/strong><\/h4>\n<p>Leptin increases the <a href=\"https:\/\/selfhacked.com\/blog\/factors-counteract-stress-response\/\">stress response<\/a> and the activity of the sympathetic nervous system (fight or flight) [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3905446\/\">R<\/a>].<\/p>\n<p>It activates the<a href=\"https:\/\/selfhacked.com\/blog\/reasons-cortisol-low-high\/\"> HPA axis<\/a> and lowers <a href=\"https:\/\/selfhacked.com\/blog\/resting-heart-rate\/\">heart rate<\/a> variability (HRV) [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3042594\/\">R<\/a>].<\/p>\n<h4><a id=\"post-77608-_y7912jafmzow\"><\/a><strong>7) Cancer<\/strong><\/h4>\n<p>Leptin promotes the growth and spread of cancer. This is one reason why obesity is considered a risk factor for cancer [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/15042602\">R<\/a>].<\/p>\n<p>It increases angiogenesis by increasing vascular endothelial growth factor (<a href=\"http:\/\/selfhacked.com\/blog\/need-know-vegf-good-bad\/\">VEGF<\/a>) levels.<\/p>\n<h4><a id=\"post-77608-_peirrvyhvd8n\"><\/a><strong>8) Heart Disease<\/strong><\/h4>\n<p>Leptin receptors are found in the heart [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/15749744\">R<\/a>]. Studies have linked high leptin levels to an increased risk of cardiovascular disease [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/15749744\">R<\/a>].<\/p>\n<p>In obese subjects, leptin levels are the most significant predictor of higher levels of <a href=\"https:\/\/selfhacked.com\/blog\/fibrinogen\/\">fibrinogen<\/a>, a clotting factor considered to be one of the most important risk factors for heart disease [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/12135695\">R<\/a>].<\/p>\n<p>Even so, high leptin levels also are associated with high blood pressure [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/16404481\">R<\/a>], although this might be because of leptin resistance because leptin infusion in rats lowers blood pressure [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/20059648\">R<\/a>].<\/p>\n<p>Leptin increases <a href=\"https:\/\/selfhacked.com\/blog\/endothelins\/\">endothelin<\/a>-1 production, which is a potent vasoconstrictor [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/16404481\">R<\/a>]. By constricting blood vessels, leptin\/endothelin-1 could play a role in the development of heart disease [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/16404481\">R<\/a>].<\/p>\n<h4><a id=\"post-77608-_5e5rw8u629q9\"><\/a><strong>9) Pain Sensitivity<\/strong><\/h4>\n<p>Higher leptin levels in the body can cause the body&#8217;s<a href=\"https:\/\/selfhacked.com\/blog\/pain\/\"> pain<\/a> sensitivity to increase. This is because it can cause inflammation, which plays a role in pain [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4939369\/\">R<\/a>].<\/p>\n<p>In a study of 3 fibromyalgia patients, they reported higher pain levels on days when they had increased leptin in their blood. A cohort study of 5,600 postmenopausal women showed similar results [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4939369\/\">R<\/a>].<\/p>\n<p>Leptin may increase pain by activating inflammatory markers (<a href=\"http:\/\/selfhacked.com\/blog\/interleukin-1\/\">IL-1B<\/a>, IL-6, <a href=\"http:\/\/selfdecode.com\/gene\/IL18\/?utm_source=seo&#038;utm_medium=selfhacked&#038;utm_campaign=id00002\">IL-18<\/a>) or preventing pain modulating signaling [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4939369\/\">R<\/a>].<\/p>\n<h4><a id=\"post-77608-_dq7lakdl4kk2\"><\/a><strong>10) Nutrient Absorption<\/strong><\/h4>\n<p>It is secreted by the stomach and enters the small intestine. Several studies have shown that leptin interferes with the absorption of some nutrients.<\/p>\n<p>It enhances <a href=\"http:\/\/selfhacked.com\/blog\/health-benefits-butyrate-derivatives-sodium-butyrate-phenylbutyrate-trybutyrine-butyric-acid-butyrate-prodrugs-butyrate-producing-bacteria\/\">butyrate<\/a> uptake [<a href=\"http:\/\/journals.plos.org\/plosone\/article?id=10.1371\/journal.pone.0083360#B18\">R<\/a>], intestinal transport of fructose [<a href=\"http:\/\/journals.plos.org\/plosone\/article?id=10.1371\/journal.pone.0083360#B20\">R<\/a>] and oligopeptides [<a href=\"http:\/\/journals.plos.org\/plosone\/article?id=10.1371\/journal.pone.0083360#B21\">R<\/a>].<\/p>\n<p>Leptin decreases <a href=\"https:\/\/selfhacked.com\/blog\/glucose\/\">glucose<\/a> [<a href=\"http:\/\/journals.plos.org\/plosone\/article?id=10.1371\/journal.pone.0083360\">R<\/a>] and galactose uptake [<a href=\"http:\/\/journals.plos.org\/plosone\/article?id=10.1371\/journal.pone.0083360#B22\">R<\/a>]. It also inhibited the absorption of Proline, <a href=\"http:\/\/selfhacked.com\/blog\/beta-alanine\/\">Beta-Alanine<\/a>, and <a href=\"https:\/\/selfhacked.com\/blog\/15-proven-health-benefits-glutamine\/\">Glutamine<\/a> [<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/25349247\">R<\/a>].<\/p>\n<p>On the one hand, I could see this being both bad and good.
