It stimulates the metabolism, improves insulin sensitivity, decreases inflammation, and improves muscle performance. AMPK is also involved in several longevity pathways and may promote healthy aging.<!--more--><\/p>\n<h2><span id=\"AMPK_The_Energy_Enzyme\">AMPK: The Energy Enzyme<\/span><\/h2>\n<p><b>AMPK<\/b> (<a href=\"http:\/\/selfhacked.com\/blog\/natural-ampk-activators\/\">5\u2032 AMP-activated protein kinase<\/a>) is an enzyme that <b>plays a key role in energy balance<\/b>. All creatures from yeast to humans have this enzyme (<a href=\"http:\/\/en.wikipedia.org\/wiki\/AMP-activated_protein_kinase\">R<\/a>).<\/p>\n<p><b>AMPK can detect the level of energy (number of ATP molecules) in a cell and helps regulate responses when it gets too low or high<\/b>.<\/p>\n<p>AMPK is produced in a number of tissues, including the liver, brain, fat cells and muscle (<a href=\"http:\/\/en.wikipedia.org\/wiki\/AMP-activated_protein_kinase\">R<\/a>).<\/p>\n<p>While much of AMPK activity is dependent on external factors such as diet and <a href=\"https:\/\/selfhacked.com\/blog\/top-14-proven-health-benefits-exercise-references-mechanisms\/\">exercise<\/a> <b>we all have a genetic disposition inherited from our parents<\/b>.<\/p>\n<h2><span id=\"Health_Effects_of_AMPK_Activation\"><b>Health Effects of AMPK Activation<\/b><\/span><\/h2>\n<h3><span id=\"1_Increases_Metabolism\"><b>1) Increases Metabolism<\/b><\/span><\/h3>\n<p>AMPK in the<a href=\"https:\/\/selfhacked.com\/blog\/hypothalamus-101\/\"> hypothalamus<\/a> senses our level of energy production in the body (in the form of ATP). It increases energy expenditure and can also increase appetite (when it is activated in the hypothalamus) (<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4980534\/\">R<\/a>).<\/p>\n<p>When cellular energy is low, AMPK is activated and targets a range of processes, the net response of which is an increase in energy production and a coordinated decrease in energy (ATP) usage (<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4287273\/\">R<\/a>).<\/p>\n<p>Hypothalamic AMPK increases appetite, increases <a href=\"https:\/\/selfhacked.com\/blog\/glucose\/\">glucose<\/a> production and uptake, reduces heat production, and decreases energy output (<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4980534\/\">R<\/a>).<\/p>\n<h3><span id=\"2_Produces_and_Burns_Sugars\"><b>2) Produces and Burns Sugars<\/b><\/span><\/h3>\n<p><a href=\"https:\/\/selfhacked.com\/blog\/glucose\/?utm_source=seo&amp;utm_medium=selfhacked&amp;utm_campaign=id00002\">Glucose<\/a> is the main source of energy for the body and is particularly essential for normal brain activity. Hypoglycemia, a condition in which the blood glucose drops below normal levels, poses a great danger to the stability and functioning of the brain and therefore activates AMPK (<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4980534\/\">R<\/a>).<\/p>\n<p>Hypothalamic AMPK activation <b>promotes glucose production from the liver<\/b> (<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4980534\/\">R<\/a>) and <b>glucose uptake into the muscles <\/b>(<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4973318\/\">R<\/a>).<\/p>\n<p>AMPK inhibits glucose storage (glycogen synthesis), resulting in more glucose being available for energy production (<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4973318\/\">R<\/a>).<\/p>\n<p>In various cells, AMPK <b>stimulates the breakdown of glucose for energy (in the form of ATP)<\/b> (<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4973318\/\">R<\/a>).<\/p>\n<h3><span id=\"3_Burns_Fat\"><b>3) Burns Fat<\/b><\/span><\/h3>\n<p>AMPK inhibits the production of fatty acids, <a href=\"https:\/\/selfhacked.com\/blog\/cholesterol\/\">cholesterol<\/a>, and <a href=\"https:\/\/selfhacked.com\/blog\/triglycerides\/\">triglycerides<\/a>, and instead stimulates the breakdown and burning of existing fat for energy (<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4973318\/\">R<\/a>).<\/p>\n<h3><span id=\"4_Inhibits_Protein_Production\"><b>4) Inhibits Protein Production<\/b><\/span><\/h3>\n<p>Protein production is a high-energy process that is inhibited during low energy states to conserve energy. Therefore, it is not surprising that <b>AMPK inhibits protein production<\/b> (<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4973318\/\">R<\/a>).<\/p>\n<p>Inhibiting excessive protein production results in a much more energy-efficient and less wasteful cell.<\/p>\n<h3><span id=\"5_Promotes_Cellular_Recycling_Autophagy\"><b>5) Promotes Cellular Recycling (Autophagy)<\/b><\/span><\/h3>\n<p><a href=\"https:\/\/selfhacked.com\/blog\/everything-wanted-know-autophagy\/\">Autophagy<\/a> is the process of recycling cellular components. However, metformin (AMPK activator) is commonly given to women with PCOS to a good effect (PCOS is a condition with higher male hormones); much more research is required to determine AMPK\u2019s role in testosterone production.<\/p>\n<h3><span id=\"A_Role_in_Neurodegenerative_Disease\"><b>A Role in Neurodegenerative Disease?<\/b><\/span><\/h3>\n<p><b>AMPK has both protective and contributing properties when it comes to neurodegenerative diseases<\/b>, and studies often come to opposing conclusions regarding this enzyme and its role.<\/p>\n<p>In mice with Alzheimer\u2019s, activation of AMPK by metformin was shown to increase amyloid-beta protein (A\u03b2) levels and therefore contribute to the disease (<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4287273\/\">R<\/a>).<\/p>\n<p>However, A\u03b2 generation was also shown to be increased in AMPK deficient mice (<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4864506\/\">R<\/a>). Furthermore, AMPK activation by<a href=\"https:\/\/selfhacked.com\/blog\/top-15-scientific-health-benefits-of-resveratrol-with-references\/\"> resveratrol<\/a> and AICAR also decreased A\u03b2 secretion (<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4287273\/\">R<\/a>).<\/p>\n<p>Indeed, the inactivation of AMPK was linked to increased Alzheimer\u2019s risk in obese patients with type 2 diabetes (<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4287273\/\">R<\/a>).<\/p>\n<p>In mice with Huntington\u2019s disease, AMPK activation promoted neuronal loss and brain decay (<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4287273\/\">R<\/a>). But then again, another study showed that treatment with metformin significantly prolonged survival time in rodents with this disease (<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4101001\/\">R<\/a>).<\/p>\n<p>Genetic activation of AMPK was also shown to protect against neuronal loss in Parkinson\u2019s disease models in flies (<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4287273\/\">R<\/a>).<\/p>\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Overactivation or underactivation of AMPK may each contribute to neurodegenerative disease, and the safest bet is to lead a healthy and balanced life. More research is needed.<\/div><\/div>\n<h3><span id=\"A_Role_in_Cancer\"><b>A Role in Cancer?<\/b><\/span><\/h3>\n<p><b>The role of AMPK in cancer is complicated and poorly understood<\/b> (<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4973318\/\">R<\/a>).<\/p>\n<p>AMPK activation can protect against DNA damage from<a href=\"https:\/\/selfhacked.com\/blog\/oxidative-stress-101\/\"> oxidative stress<\/a>.
