It was discovered while searching for molecules that imitate the function of brain-derived neurotrophic factor (<a href=\"http:\/\/selfhacked.com\/blog\/a-comprehensive-list-of-natural-ways-to-increase-bdnf\/\">BDNF<\/a>) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC5319242\/\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4702337\/\"> R<\/a>].<\/p>\n<p>BDNF promotes the growth of neurons and synapses (synaptogenesis) and is very important for normal brain function. Lower amounts of BDNF are observed in diseases such as <a href=\"https:\/\/selfhacked.com\/blog\/natural-treatments-depression\/\">depression<\/a>, Alzheimer\u2019s, Parkinson\u2019s, and schizophrenia [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC5319242\/\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC5295364\/\"> R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4828984\/\"> R<\/a>].<\/p>\n<p>Studies in animals show that 7,8-DHF could potentially help with brain repair, long-term memory, depression, and neurodegenerative diseases. However, studies in humans have not yet begun [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4702337\/\">R<\/a>].<\/p>\n<h3><span id=\"Mechanism_of_Effect\"><a id=\"post-43181-_o37hnqdllga7\"><\/a><strong>Mechanism of Effect<\/strong><\/span><\/h3>\n<p>7,8-DHF<strong> mimics the effects of brain-derived neurotrophic factor (<\/strong><a href=\"https:\/\/selfhacked.com\/blog\/a-comprehensive-list-of-natural-ways-to-increase-bdnf\/\"><strong>BDNF<\/strong><\/a><strong>)<\/strong> in brain cells by activating tropomyosin-related kinase B (<a href=\"http:\/\/selfdecode.com\/gene\/ntrk2\/?utm_source=seo&amp;#38;utm_medium=selfhacked&amp;#38;utm_campaign=id00002\">TrkB<\/a>) receptors, the typical target of BDNF [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/20133810\">R<\/a>].<\/p>\n<p>The therapeutic potential of BDNF is restricted due to its short half-life (less than 10 minutes) and its inability to cross the blood-brain barrier because of its large size. Unlike BDNF <strong>7,8-DHF<\/strong> <strong>is able to penetrate the<\/strong><a href=\"https:\/\/selfhacked.com\/blog\/blood-brain-barrier-causes-tests-leaky-brain\/\"><strong> blood-brain barrier<\/strong><\/a> and enter the central nervous system (CNS) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC5319242\/\">R<\/a>].<\/p>\n<p>7,8-DHF also increases the production of <a href=\"http:\/\/selfhacked.com\/blog\/about-nrf2-and-natural-ways-to-increase-it\/\">Nrf2<\/a>. Nrf2 increases antioxidants enzymes such as heme oxygenase 1 (<a href=\"https:\/\/selfdecode.com\/gene\/HMOX1\/?utm_source=seo&amp;#38;utm_medium=selfhacked&amp;#38;utm_campaign=id00002\">HO-1<\/a>) and also enzymes that repair DNA (8-oxoguanine DNA glycosylase-1 &#8211; OGG1) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24610348\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24151624\"> R<\/a>].<\/p>\n<h4><a id=\"post-43181-_c62ifae3ii9i\"><\/a>Antioxidant Activity<\/h4>\n<p>7,8-DHF rescues cells from damage and death caused by<a href=\"https:\/\/selfhacked.com\/blog\/oxidative-stress-101\/\"> oxidative stress<\/a> [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24220540\">R<\/a>].<\/p>\n<p>Cells do not need to have the TrkB receptor to be protected [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24220540\">R<\/a>].<\/p>\n<p>In this case, 7,8-DHF:<\/p>\n<ul>\n<li>Removed reactive oxygen species (<a href=\"https:\/\/selfhacked.com\/blog\/oxidative-stress-101\/\">ROS<\/a>) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/19182370\">R<\/a>].<\/li>\n<li>Increased<a href=\"https:\/\/selfhacked.com\/blog\/about-nrf2-and-natural-ways-to-increase-it\/\"> Nrf2<\/a>. This increases the production of several antioxidant enzymes [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24610348\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24151624\"> R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24503931\"> R<\/a>].<\/li>\n<li>Increased <a href=\"http:\/\/selfhacked.com\/blog\/30-scientifically-proven-health-benefits-glutathione\/\">glutathione<\/a> (<a href=\"http:\/\/selfhacked.com\/blog\/30-scientifically-proven-health-benefits-glutathione\/\">GSH<\/a>), <a href=\"https:\/\/selfdecode.com\/gene\/gpx1\/?utm_source=seo&amp;#38;utm_medium=selfhacked&amp;#38;utm_campaign=id00002\">glutathione peroxidase<\/a> (<a href=\"https:\/\/selfdecode.com\/gene\/gpx1\/?utm_source=seo&amp;#38;utm_medium=selfhacked&amp;#38;utm_campaign=id00002\">GPx<\/a>), and superoxide dismutase (<a href=\"http:\/\/selfhacked.com\/blog\/the-brain-fog-gene-rs4880-or-sod2\/\">SOD<\/a>) levels [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24045895\">R<\/a>].<\/li>\n<\/ul>\n<h3><span id=\"R13_in_Alzheimers_Disease\"><a id=\"post-43181-_sxia1dlulub\"><\/a>R13 in Alzheimer\u2019s Disease<\/span><\/h3>\n<p>Because of the relatively low apparent bioavailability of 7,8-DHF (about 5% in mice), researchers are currently developing prodrugs that can be converted to 7,8-DHF once inside the body. The most promising of these is currently known as R13, which has eliminated plaques associated with Alzheimer\u2019s disease in the brains of living mice [<a href=\"https:\/\/patents.google.com\/patent\/US20150274692\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC5777001\/\">R<\/a>].<\/p>\n<p>R13 has not yet been tested in humans, and we do not recommend using it until such studies are conducted.<\/p>\n<h2><span id=\"Animal_Cell_Research_on_78-Dihydroxyflavone\"><a id=\"post-43181-_yfok7821gcpd\"><\/a><strong>Animal &amp; Cell Research on 7,8-Dihydroxyflavone<\/strong><\/span><\/h2>\n<p><strong>No clinical evidence supports the use of 7,8,-DHP for any health condition<\/strong>, as all research thus far has been preclinical. Below is a summary of the existing animal and cell-based research, which should guide further investigational efforts. However, the studies listed below should not be interpreted as supportive of any health benefit.<\/p>\n<h3><span id=\"1_Memory_and_Learning\"><a id=\"post-43181-_ie5bsxxspxl\"><\/a>1) Memory and Learning<\/span><\/h3>\n<p>7,8-DHF improved object recognition (a test used to determine learning and memory) in healthy rats when given immediately following and three hours after learning. The beneficial effect was more pronounced in females [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24045895\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/23469848\"> R<\/a>].<\/p>\n<p>7,8-DHF also prevented neuronal damage in mice after traumatic brain injury [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/25801526\">R<\/a>].<\/p>\n<h3><span id=\"4_Inflammation\"><a id=\"post-43181-_y2fegseuihu\"><\/a>4) Inflammation<\/span><\/h3>\n<p>7,8-DHF decreased the release of inflammatory factors in brain cells by blocking<a href=\"https:\/\/selfhacked.com\/blog\/nuclear-factor-kappa-b\/\"> NF-\u03baB<\/a> [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24535427\">R<\/a>].<\/p>\n<p>7,8-DHF also reduced the levels of inflammation-causing nitric oxide, <a href=\"https:\/\/selfhacked.com\/blog\/prostaglandins\/\">prostaglandin<\/a> E2 (<a href=\"http:\/\/selfdecode.com\/gene\/ptges2\/?utm_source=seo&amp;#38;utm_medium=selfhacked&amp;#38;utm_campaign=id00002\">PGE2<\/a>), <a href=\"http:\/\/selfhacked.com\/blog\/supplements-lifestyle-factors-influence-tnf-interleukin-6-il-6\/\">TNF-alpha<\/a>, and <a href=\"http:\/\/selfhacked.com\/blog\/interleukin-6\/\">IL-6<\/a> in macrophages (<a href=\"https:\/\/selfhacked.com\/blog\/how-to-increase-and-decrease-white-blood-cells\/\">white blood cells<\/a>) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/28295316\">R<\/a>].<\/p>\n<h3><span id=\"5_Neurodegenerative_Disease\"><a id=\"post-43181-_sx0p5g5b2d6t\"><\/a>5) Neurodegenerative Disease<\/span><\/h3>\n<h4><a id=\"post-43181-_bcpgpgpnjtk1\"><\/a><strong>Alzheimer\u2019s Disease<\/strong><\/h4>\n<p>In animal models for<a href=\"https:\/\/selfhacked.com\/blog\/alzheimer\/\"> Alzheimer\u2019s<\/a> disease, 7,8-DHF [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24325271\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24022672\"> R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24614170\"> R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24070857\"> R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/26577931\"> R<\/a>]:<\/p>\n<ul>\n<li>Reduced amyloid plaque formation<\/li>\n<li>Reduced oxidative stress<\/li>\n<li>Prevented loss of synapses<\/li>\n<li>Prevented memory deficits and preserved cognitive function<\/li>\n<\/ul>\n<p>However, another study found no benefits in treating mice with Alzheimer\u2019s-like brain damage with 7,8-DHF [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/25523427\">R<\/a>].<\/p>\n<h4><a id=\"post-43181-_e6xe6hfg0rg8\"><\/a><strong>Parkinson\u2019s Disease<\/strong><\/h4>\n<p>7,8-DHF improved motor function and prevents the loss of<a href=\"https:\/\/selfhacked.com\/blog\/dopamine\/\"> dopamine<\/a>-related neurons in a mouse model of Parkinson\u2019s disease [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/25655214\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/27079181\"> R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/27019033\"> R<\/a>].<\/p>\n<p>It also prevents the death of dopamine-sensitive neurons in monkey models of Parkinson\u2019s disease [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/27731318\">R<\/a>].<\/p>\n<h4><a id=\"post-43181-_3mno80dc8qf1\"><\/a><strong>Huntington\u2019s Disease<\/strong><\/h4>\n<p>7,8-DHF delayed the motor and cognitive impairment and prolonged survival in a mouse model of Huntington\u2019s disease [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/23446639\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/28541476\"> R<\/a>].<\/p>\n<h4><a id=\"post-43181-_rt1xoqe42v1\"><\/a><strong>Amyotrophic Lateral Sclerosis (ALS)<\/strong><\/h4>\n<p>7,8-DHF improved motor deficits and neuron survival in a mouse model of ALS [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24637017\">R<\/a>].<\/p>\n<h4><a id=\"post-43181-_trqibvuaicxz\"><\/a><strong>Multiple Sclerosis<\/strong><\/h4>\n<p>7,8-DHF reduced disease severity in a mouse model of<a href=\"https:\/\/selfhacked.com\/blog\/causes-symptoms-diagnosis-risk-factors-of-multiple-sclerosis\/\"> multiple sclerosis<\/a> [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/26943953\">R<\/a>].<\/p>\n<h4><a id=\"post-43181-_7pbww866s8yt\"><\/a><strong>Schizophrenia<\/strong><\/h4>\n<p>7,8-DHF decreased cognitive deficits and improved learning and memory in rat models of schizophrenia [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24662915\">R<\/a>].<\/p>\n<p>Infections in pregnancy and subsequent abnormal brain development can increase the risk of schizophrenia in offspring. Early use of 7,8-DHF decreased behavioral abnormalities and psychosis in mice offspring at risk of developing a schizophrenia-like disorder [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/28138113\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/27824119\"> R<\/a>].<\/p>\n<h4><a id=\"post-43181-_3cunhs4lja9j\"><\/a><strong>Down Syndrome<\/strong><\/h4>\n<p>In a mouse model of down syndrome, early intervention with 7,8-DHF increased the production of new neurons (hippocampus) and improved learning and memory [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/28882412\">R<\/a>].<\/p>\n<h4><a id=\"post-43181-_etvfoghfgpzp\"><\/a><strong>Fragile X Syndrome<\/strong><\/h4>\n<p>Fragile X syndrome is a genetic condition. It causes a range of developmental problems including cognitive impairment and learning disabilities.<\/p>\n<p>In a mouse model of fragile X syndrome, 7,8-DHF improved cognitive function and reduced spine abnormalities [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/25229717\">R<\/a>].<\/p>\n<h4><a id=\"post-43181-_gz9gkee25fib\"><\/a><strong>Rett Syndrome<\/strong><\/h4>\n<p>Rett Syndrome is a non-inherited genetic brain disorder, mainly affecting girls.
