It&#8217;s important to try and figure out whether you are a high or low PXR producer &#8211; and we can figure that out by genes, symptoms and blood tests.<\/p>\n<p>Read more below to learn about PXR.<\/p>\n<p><!--more--><\/p>\n<h2><span id=\"What_is_the_Pregnane_X_Receptor\">What is the Pregnane X Receptor?<\/span><\/h2>\n<p>The pregnane X receptor (PXR) is implicated in fatty liver, vitamin D homeostasis, bile acids homeostasis, steroid hormones homeostasis and inflammatory bowel diseases (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2593625\/\">R<\/a>).<\/p>\n<p>The pregnane X receptor (PXR) has a significant role in how the body handles any foreign substances, especially manmade chemicals (<a href=\"http:\/\/www.nature.com\/articles\/srep31936\">R<\/a>).<\/p>\n<p>PXR is one of several cell receptors that are activated by components of the liver\u2019s bile (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/27459945\">R<\/a>).<\/p>\n<p>In newborn mice, exposure to PXR activators directed the genes responsible for the metabolism of extra compounds and fats (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/27413110\">R<\/a>).<\/p>\n<p>Enzymes produced by PXR (such as CYP3A4) that help us detox are reduced in inflammatory states (<a href=\"http:\/\/dmd.aspetjournals.org\/content\/35\/9\/1687.full\">R<\/a>).<\/p>\n<p>This is one reason why people suffering from chronic inflammation often feel like they are reacting to all drugs that they take.<\/p>\n<h2><span id=\"Why_Do_You_Not_Feel_Same_Effects_The_First_Time_You_Take_a_Drug\">Why Do You Not Feel Same Effects The First Time You Take a Drug?<\/span><\/h2>\n<p>PXR can explain why you don&#8217;t feel the same effects from a drug\/herb if you take it daily.<\/p>\n<p>When you take a drug that activates PXR (many do), then it will produce a bunch of enzymes that can eliminate the drug quite effectively (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2651806\/\">R<\/a>), which is probably why you won&#8217;t notice the same effects from the same dosage.<\/p>\n<h2><span id=\"Pros_of_PXR\">Pros of PXR<\/span><\/h2>\n<h3><span id=\"1_Helps_Clear_Toxins\">1) Helps Clear Toxins<\/span><\/h3>\n<p>PXR functions as a defense mechanism against toxic insults (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2651806\/\">R<\/a>).<\/p>\n<p>PXR induces genes that help with all three stages of detoxification.<\/p>\n<p>PXR increases CYP3A4, an important phase I oxidative enzyme that is responsible for the metabolism of many drugs.<\/p>\n<p>In addition, PXR increases phase II enzymes such as glutathione S-transferase and phase III transport uptake and efflux proteins such as OATP2 and MDR1 (<a href=\"https:\/\/en.wikipedia.org\/wiki\/Pregnane_X_receptor\">R<\/a>).<\/p>\n<h3><span id=\"2_Decreases_Inflammation\">2) Decreases Inflammation<\/span><\/h3>\n<p>PXR is responsible for <strong>decreasing the amount of inflammation<\/strong> that occurs during infection in mice (<a href=\"http:\/\/www.nature.com\/articles\/srep31936\">R<\/a>).<\/p>\n<p>By modifying the concentration of various enzymes within the intestinal lining, PXR is responsible for maintaining the gut during inflammation (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/27440420\">R<\/a>).<\/p>\n<p id=\"P20\" class=\"p p-first\">Activation of PXR in humans inhibits the activity of NF-\u03baB, a key regulator of inflammation and the immune response. In mice without PXR, the expression of NF-\u03baB target genes is substantially increased in multiple tissues, and small bowel inflammation is significantly increased (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2593625\/\">R<\/a>).<\/p>\n<p class=\"p p-first\">Furthermore, PXR-deficient lymphocytes produce more IFN-gamma and less of the anti-inflammatory cytokine IL-10 (<a href=\"http:\/\/www.jimmunol.org\/content\/184\/6\/2949.full.pdf\">R<\/a>), which suggests that PXR suppresses Th1 inflammation.<\/p>\n<h3 id=\"P21\" class=\"p p-last\"><span id=\"3_Helps_Liver_Function\">3) Helps Liver Function<\/span><\/h3>\n<p>Fetus exposure to PXR-activators <strong>helps regulate gene activity related to lipid metabolism in the liver<\/strong> (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/27413110\">R<\/a>).<\/p>\n<p>Using humans, a mouse model, and cultured cells, scientists found a connection between the PXR pathway and decreased the activity of key enzymes involved in nonalcoholic fatty liver disease (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/27482056\">R<\/a>).<\/p>\n<p>PXR-mediated lipid accumulation is required for liver regeneration [<a id=\"__tag_160504240\" class=\"bibr popnode tag_hotlink tag_tooltip\" href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2593625\/#R58\">R<\/a>], suggesting that PXR is essential for normal progression of liver regeneration by modulating lipid homeostasis.<\/p>\n<h3><span id=\"4_Helps_Detox_Bile_Bilirubin\">4) Helps Detox Bile, Bilirubin<\/span><\/h3>\n<p>PXR helps detox products such as bile and bilirubin (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/15986414\">R<\/a>), so if you have Gilbert&#8217;s Syndrome, you may not have enough PXR.<\/p>\n<p>Removing bile can help prevent cholestasis (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/15986414\">R<\/a>).<\/p>\n<p>PXR has been shown to induce the expression of multiple key components in the clearance pathway of bile and bilirubin, including UGT1A1, OATP2, GSTA1 and 2 and MRP2 (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2593625\/\">R<\/a>).<\/p>\n<p>Consistent with the pattern of gene regulation, activation of PXR in mice has been shown to prevent experimental hyperbilirubinemia [<a id=\"__tag_160504271\" class=\"bibr popnode tag_hotlink tag_tooltip\" href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2593625\/#R53\">R<\/a>].<\/p>\n<aside class=\"sh-partner shp-form sponsored sh-et-7388fa\">\n  <div>\n          <div class=\"partner-heading\">8 Ways To Use Your DNA &amp; Optimize Your Health<\/div>\n    \n    <div class=\"partner-content\">\n      <p><span style=\"font-weight: 400\">What kinds of secrets could your genes hold? PXR may work by increasing SCD1 and increased the production of unsaturated fatty acids (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2593625\/\">R<\/a>).<\/p>\n<h2><span id=\"Cons_of_PXR\">Cons of PXR<\/span><\/h2>\n<h3><span id=\"1_PXR_Can_Inhibit_Anti-Cancer_Medication\">1) PXR Can Inhibit Anti-Cancer Medication<\/span><\/h3>\n<p>PXR plays a role in the genes that oversee drug resistance. This can <strong>complicate treatment in some forms of cancer<\/strong> (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/27572875\">R<\/a>).<\/p>\n<p>Suppressing PXR by way of inhibitory drugs can enhance the effect of anticancer medications for ovarian cancer. This helps the body overcome the treatment difficulties brought on by drug resistance (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/27572875\">R<\/a>).<\/p>\n<p>In human colon cancer cells, PXR was shown to inhibit a crucial tumor-suppressing gene. This can <strong>cause tumors to grow bigger<\/strong> (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4979430\/\">R<\/a>).<\/p>\n<p>PXR can increase the recurrence of colon cancer tumor cells. It also stops programmed cell death in colon cancer cells (<a href=\"http:\/\/www.impactjournals.com\/oncotarget\/index.php?journal=oncotarget&amp;page=article&amp;op=view&amp;path%5b%5d=10646&amp;pubmed-linkout=1\">R<\/a>).<\/p>\n<h3><span id=\"2_Activation_of_PXR_Can_Interfere_With_Drug_Metabolism\">2) Activation of PXR Can Interfere With Drug Metabolism<\/span><\/h3>\n<p>Chemicals in industrialized areas that produce plastic can be potentially toxic activators of the pregnane X receptor (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/27551952\">R<\/a>).<\/p>\n<p>New brominated flame retardants and their byproducts activate PXR. This possibly causes <strong>undesirable interactions between drugs<\/strong>,<strong> interfering with the normal metabolism<\/strong>,<strong> or the body<\/strong>\u2019<strong>s ability to maintain healthy hormone levels<\/strong> (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/27506419\">R<\/a>).<\/p>\n<p>PXR is sensitive to metabolites produced by microbes, especially those that infect the intestine (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/27440420\">R<\/a>).<\/p>\n<h3><span id=\"3_PXR_Reduces_Vitamin_D\">3) PXR Reduces Vitamin D<\/span><\/h3>\n<p>Vitamin D is essential for the maintenance of calcium homeostasis and for the development and maintenance of bones (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2593625\/\">R<\/a>).<\/p>\n<p>PXR increases two enzymes (CYP24, CYP3A4) that break down 1,25 D<sub>3<\/sub>\/Calcitriol, the active form of vitamin D in humans (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2593625\/\">R<\/a>).<\/p>\n<p>This suggests that activation of PXR by some drugs may be responsible for the acceleration of vitamin D breakdown through increasing CYP3A4 and CYP24, leading to vitamin D deficiency and, eventually, to bone loss (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2593625\/\">R<\/a>).<\/p>\n<h3 class=\"inline\"><span id=\"4_PXR_Decreases_Fat_Burning_and_Inhibits_Ketogenesis\">4) PXR Decreases Fat Burning and Inhibits Ketogenesis<\/span><\/h3>\n<p id=\"P19\" class=\"p p-first-last\">Activation of PXR in the livers of mice resulted in an increased deposit of triglycerides in the liver (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2593625\/\">R<\/a>).<\/p>\n<p class=\"p p-first-last\">The activation of PXR was also associated with an inhibition of pro-fat burning genes, such as PPARa, thiolase, Cpt1a and Hmgcs2 (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2593625\/\">R<\/a>).<\/p>\n<p class=\"p p-first-last\">On the other hand, it increases the fat-storage\/increasing genes PPARy and Scd1 (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2593625\/\">R<\/a>).<\/p>\n<p class=\"p p-first-last\">Thus, activated PXR increasing triglyceride synthesis, while decreasing \u03b2-oxidation and ketogenesis in the fasting mice (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2593625\/\">R<\/a>).<\/p>\n<h3 class=\"inline\"><span id=\"5_PXR_Increases_Adrenal_Hormones_CortisolAldosterone\">5) PXR Increases Adrenal Hormones (Cortisol\/Aldosterone)<\/span><\/h3>\n<p id=\"P17\" class=\"p p-first\">Activation of PXR in mice markedly increased plasma concentrations of cortisol and aldosterone [<a class=\"bibr popnode\" href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2593625\/#R55\">R<\/a>].<\/p>\n<p class=\"p p-first\">The increased levels of corticosterone and aldosterone were associated with activation of adrenal enzymes, including <em>Cyp11a1<\/em>, <em>Cyp11b1<\/em>, <em>Cyp11b2<\/em> and <em>3<\/em>\u03b2-<em>Hsd <\/em>[<a class=\"bibr popnode\" href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2593625\/#R55\">R<\/a>].<\/p>\n<p class=\"p p-first\">The PXR-activating mice also exhibited overgrowth of the adrenal cortex, loss of cortisol circadian rhythm, and a lack of cortisol responses to psychological stress [<a class=\"bibr popnode\" href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2593625\/#R55\">R<\/a>].<\/p>\n<p class=\"p p-first\">It could contribute to Pseudo-Cushing&#8217;s syndrome, which is most seen in alcoholic, depressed or obese subjects [<a class=\"bibr popnode\" href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2593625\/#R55\">R<\/a>].<\/p>\n<h3 class=\"p p-first\"><span id=\"6_PXR_and_Colon_Cancer\">6) PXR and Colon Cancer<\/span><\/h3>\n<p>PXR may promote colon cancer in a certain model by inhibiting <a href=\"https:\/\/selfdecode.com\/gene\/tp53\/\">p53<\/a>, a tumor suppressor gene (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4979430\/\">R<\/a>).<\/p>\n<h3><span id=\"7_PXR_and_Oxidative_Stress_Responses\">7) PXR and Oxidative Stress Responses<\/span><\/h3>\n<p>PXR seems to have mixed effects on oxidative stress (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/16195250\/\">R<\/a>).<\/p>\n<p>Activated PXR in transgenic female mice resulted in a heightened sensitivity to an oxidative toxicant (<a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/16195250\/\">R<\/a>).<\/p>\n<p>The PXR-induced sensitivity was associated with decreased activities of superoxide dismutase and <a href=\"https:\/\/selfhacked.com\/blog\/catalase-benefits\/\">catalase<\/a>, enzymes that scavenge superoxide and hydrogen peroxide, respectively (<a
