In a study of 17 healthy volunteers who underwent brain imaging, uridine supplements increased their brain levels of phosphoethanolamine (PEtn), an important phospholipid building block [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3020593\/\">R<\/a>].<\/p>\n<p><b>Uridine creates the right environment for increased neuronal plasticity and synapse formation, which may help the brain adapt, learn, remember, and regrow<\/b> [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/15939540\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/19262950\"> R<\/a>].<\/p>\n<p>In cell and animal studies, uridine increased <i>neurite outgrowth<\/i>, a process that helps brain cells grow and regenerate [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/15939540\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/19262950\"> R<\/a>].<\/p>\n<p>In the same studies, uridine also increased synaptic proteins<b>,<\/b> small molecules found in the cleft or synapse that brain cells use to communicate. When these proteins drop, synapses get destroyed. Only later do brain cells die and memory problems arise, as in Alzheimer\u2019s disease [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/15939540\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/19262950\"> R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/30606734\"> R<\/a>].<\/p>\n<p>Lastly, uridine enhanced dendritic spines, the &#8220;back&#8221; of neurons thought to store memory [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/15939540\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/19262950\"> R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4573337\/\"> R<\/a>].<\/p>\n<p>Two gerbil studies came to a similar conclusion. In one, uridine monophosphate in combination with<a href=\"https:\/\/selfhacked.com\/blog\/top-18-scientific-health-benefits-choline-cdp-choline-alpha-gpc\/\"> choline<\/a> and <a href=\"http:\/\/selfhacked.com\/blog\/top-22-science-based-health-benefits-of-fish-oil\/\">DHA<\/a> enhanced cognitive function. In the other, uridine and DHA increased phospholipids and synaptic protein levels in the animals&#8217; brains [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/16631143\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/18606862\"> R<\/a>].<\/p>\n<p>Uridine activates the <a href=\"https:\/\/selfdecode.com\/gene\/P2RY2\/?utm_source=seo&amp;#38;utm_medium=selfhacked&amp;#38;utm_campaign=id00002\">P2Y2<\/a><a href=\"https:\/\/selfdecode.com\/gene\/P2RY2\/\"> receptor<\/a>, which, alongside<a href=\"https:\/\/selfhacked.com\/blog\/all-about-nerve-growth-factor-and-50-ways-to-increase-it\/\"> nerve growth factor<\/a> (<a href=\"http:\/\/selfhacked.com\/blog\/all-about-nerve-growth-factor-and-50-ways-to-increase-it\/\">NGF<\/a>), promotes the birth of new neurons and regeneration of injured ones [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/23550835\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC1323158\/\"> R<\/a>].<\/p>\n<p>An active P2Y2 receptor likely protects neurons from injury in the first place. It becomes more active under inflammatory conditions, such as when interleukin-1b (<a href=\"http:\/\/selfhacked.com\/blog\/interleukin-1\/\">IL-1b<\/a>) is high, to protect the brain [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3360094\/\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/23550835\"> R<\/a>].<\/p>\n<p>Although promising, most of the effects of uridine on learning and memory have been observed in animals and cells. Membrane phospholipids may be altered in people with bipolar disorder [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/21176029\">R<\/a>].<\/p>\n<p>Also, in rats, uridine in combination with <a href=\"http:\/\/selfhacked.com\/blog\/top-22-science-based-health-benefits-of-fish-oil\/\">omega-3<\/a> fatty acids relieved symptoms of depression [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/15705349\">R<\/a>].<\/p>\n<p>All in all, the evidence to support the role of uridine in improving depression is insufficient. Larger, more robust clinical trials are required.<\/p>\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Preliminary research suggests that uridine may help relieve depression and bipolar disorder by improving mitochondrial function and cell membrane health.<\/div><\/div>\n<h3><span id=\"3_Nerve_Pain_Damage\"><b>3) Nerve Pain &amp; Damage<\/b><\/span><\/h3>\n<p>In 212 people with peripheral neuropathy, a combination of uridine, <a href=\"https:\/\/selfhacked.com\/blog\/16-proven-benefits-vitamin-b9-folate-folic-acid\/\">folic acid<\/a>, and<a href=\"https:\/\/selfhacked.com\/blog\/vitamin-b12\/\"> vitamin <\/a><a href=\"https:\/\/selfhacked.com\/blog\/thiamine\/\">B1<\/a><a href=\"https:\/\/selfhacked.com\/blog\/vitamin-b12\/\">2<\/a> significantly <b>lowered both the intensity of pain and the number of areas affected<\/b> [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/26679082\">R<\/a>].<\/p>\n<p>Another trial of 48 people suffering from carpal tunnel syndrome seems to confirm this finding: a similar combination lowered the intensity of the pain by about 40% [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/26679082\">R<\/a>].<\/p>\n<p>In a trial of 400 people, the combination of vitamin<a href=\"https:\/\/selfhacked.com\/blog\/vitamin-b12\/\"> B12<\/a>, uridine, and cytidine effectively reduced nerve pain with only mild side effects. The combination worked better than vitamin B12 alone [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC5317309\/\">R<\/a>].<\/p>\n<p>Uridine also improved nerve health in 20 patients with diabetic neuropathy, long-term damage to the nerves from high blood sugar [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/1325728\">R<\/a>].<\/p>\n<p>Although promising, the evidence to support the benefits of uridine in nerve pain and damage comes from a few clinical trials, most of which tested it in combination with vitamins. More clinical trials using uridine alone are needed to confirm these results.<\/p>\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Uridine, especially in combination with vitamin B12, significantly reduces nerve pain. It may also improve nerve health in people with chronic nerve damage.<\/div><\/div>\n<h3><span id=\"4_Lung_Diseases\"><b>4) Lung Diseases<\/b><\/span><\/h3>\n<p>Cystic fibrosis is a genetic disorder in which the lungs become damaged, producing a layer of sticky, persistent mucus that makes breathing a struggle.<\/p>\n<p>In 14 patients with cystic fibrosis, uridine triphosphate (UTP) and the drug amiloride <b>improved breathing and cleared mucus<\/b>; neither UTP nor amiloride alone significantly improved symptoms, while the combination restored lung function to almost normal levels [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/8665037\">R<\/a>].<\/p>\n<p>Likewise, uridine triphosphate in combination with sodium channel blockers relieved cystic fibrosis symptoms in mice [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/10678637\">R<\/a>].<\/p>\n<p>Uridine furthermore helped clear mucus in healthy people, in 15 patients with chronic mild bronchitis, and in 12 patients with primary ciliary dyskinesia (PCD), a genetic disease that prevents normal airway clearance [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/8680683\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/11463605\"> R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/10390392\"> R<\/a>].<\/p>\n<p>Uridine reduced inflammation and scarring in both animal and human cells. In the lungs of mice, it inhibited <a href=\"https:\/\/selfhacked.com\/blog\/health-benefits-collagen\/\">collagen<\/a> and <a href=\"http:\/\/selfhacked.com\/blog\/tgf\/\">TGF<\/a><a href=\"https:\/\/selfhacked.com\/blog\/tgf\/\">-\u00df<\/a> synthesis, thereby preventing the growth of scar tissue [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/26369416\">R<\/a>].<\/p>\n<p>The results are promising, but the evidence to back this use of uridine is insufficient due to the small size of the clinical trials. Further research on larger populations is warranted to confirm their results.<\/p>\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Uridine may reduce airway mucus and make it easier to breathe. In small clinical trials, it improved the symptoms of lung diseases such as cystic fibrosis and bronchitis.<\/div><\/div>\n<h3><span id=\"5_HIV-Related_Problems\"><b>5) HIV-Related Problems<\/b><\/span><\/h3>\n<p>According to one study, HIV-positive patients have lower uridine levels in their blood than healthy people do [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/21085568\">R<\/a>].<\/p>\n<p>HIV-positive patients must take antiretroviral medications to control the infection. Over the long term, these drugs cause distressing fat loss in the face, arms, buttocks, and legs [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2956768\/\">R<\/a>].<\/p>\n<p>In 165 HIV-infected patients, 24 weeks of uridine supplementation increased fat in the limbs. However, the effect wore off after 1 year. Stronger doses may have been required. Alternately, the patients may simply have been exposed to antiretroviral drugs for too long [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2956768\/\">R<\/a>].<\/p>\n<p>Long-term antiretroviral therapy is toxic to the<a href=\"https:\/\/selfhacked.com\/blog\/mitochondria\/\"> mitochondria<\/a>. One human study found uridine <b>protected against mitochondrial toxicity<\/b> from antiretrovirals. Several cell studies support this conclusion [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/16847412\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/16152713\"> R<\/a>].<\/p>\n<p>Whether uridine effectively helps prevent adverse effects of HIV medication should be confirmed in further clinical trials.<\/p>\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Blood uridine is lower in patients with HIV. Supplementation may help prevent mitochondrial toxicity and fat loss from anti-HIV drugs, but more clinical trials are needed.<\/div><\/div>\n<h3><span id=\"6_Dry_Eyes\"><b>6) Dry Eyes<\/b><\/span><\/h3>\n<p>Dry eye syndrome, which affects up to 20% of adults, causes low production of tears and inflammation of the eye surface. Oral uridine supplements <b>effectively restored the production of tears in a small trial of 27 people with dry eyes <\/b>[<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/19654956\">R<\/a>].<\/p>\n<p>A single clinical trial cannot be considered sufficient evidence to back this use. More studies on larger populations are needed before concluding that uridine helps improve dry eye syndrome.<\/p>\n<h3><span id=\"7_Sleep_Aid\"><b>7) Sleep Aid<\/b><\/span><\/h3>\n<p>Uridine is normally found in breast milk. In 30 infants with disordered sleep, a cereal enriched with uridine-5&#8242;-phosphate, tryptophan, and <a href=\"https:\/\/selfhacked.com\/blog\/adenosine-health-effects\/\">adenosine<\/a>-5&#8242;-phosphate improved<b> sleep quality. <\/b>When the infants ate the enriched cereal before bedtime, they were much more likely to sleep through the night [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/19925721\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4011061\/#__sec8title\"> R<\/a>].<\/p>\n<p>Uridine monophosphate (UMP) has been identified as a potential sleep aid in adults, but more research is required to confirm its usefulness. In rats, uridine promotes slow-wave (deep) sleep, which is the most restorative phase of sleep [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4008810\/\">R<\/a>,<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/21401495\"> R<\/a>].<\/p>\n<p>Again, very few studies have investigated the use of uridine as a sleep aid. Further clinical research is needed.<\/p>\n<h3><span id=\"Animal_and_Cell_Research_Lack_of_Evidence\"><b>Animal and Cell Research (Lack of Evidence)<\/b><\/span><\/h3>\n<p>Other potential health benefits of uridine are currently being investigated. Because the research is still at the animal and cell stage, we cannot conclude for certain that the effects will be the same in humans.<\/p>\n<h3><span id=\"Protecting_the_Heart\"><b>Protecting the Heart<\/b><\/span><\/h3>\n<p>When the heart doesn\u2019t get enough oxygen, it may suffer damage. The arteries supplying the heart are usually either partially or completely blocked, most commonly due to fatty plaques [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC1768241\/\">R<\/a>].<\/p>\n<p>In a rat study, uridine <b>normalized the heart\u2019s rhythm<\/b> <b>by reactivating energy metabolism<\/b>. It was most effective as uridine-5&#8242;-monophosphate (UMP) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/25339588\">R<\/a>].<\/p>\n<p>By activating P2Y2 receptors, uridine might also improve heart health. In a combined animal and cell study, P2Y2 receptor activation increased blood flow and improved blood pressure [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/26168216\">R<\/a>].<\/p>\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Uridine may maintain a normal heart rhythm and improve blood flow to the heart, but human studies are lacking.<\/div><\/div>\n<h3><span id=\"Liver_Regeneration\"><b>Liver Regeneration<\/b><\/span><\/h3>\n<p>People who receive liver transplants often have to take immune-suppressing drugs to prevent transplant rejection, but these drugs reduce the liver&#8217;s ability to regenerate. In rats, uridine <b>improved<\/b> <b>liver regeneration <\/b>after liver transplants. It completely restored normal liver growth [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/18713276\">R<\/a>].<\/p>\n<p>Over short periods of time, uridine prevented liver damage from drugs in animals. It seems to work as a &#8220;salvage&#8221; supplement that increases regeneration in times of high <a href=\"https:\/\/selfhacked.com\/blog\/63-factors-raising-stress-cortisol-level-backed-science\/\">stress<\/a>. The body uses it as a building block for DNA, RNA, and other important molecules [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4720477\/\">R<\/a>].<\/p>\n<p>However, chronic uridine supplementation may be harmful. In mice, uridine given to animals over a long period of time worsened liver health and triggered <a href=\"https:\/\/selfhacked.com\/blog\/insulin-101\/\">insulin<\/a><a href=\"https:\/\/selfhacked.com\/blog\/top-tips-for-fixing-insulin-resistance\/\"> resistance<\/a> [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4720477\/\">R<\/a>].<\/p>\n<p>No studies to date have looked at the effects of uridine on liver health in humans.<\/p>\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Uridine might improve liver regeneration in the short term, but it may harm the liver in the long run.
