Could taking melatonin help the body fight off COVID-19? Here&#8217;s what the most current research has to say.<\/p>\n<h3><span id=\"Mechanisms\">Mechanisms<\/span><\/h3>\n<p>The membranes of immune cells such as CD4+ T cells, <a href=\"http:\/\/selfhacked.com\/blog\/homing-fundamenal-cause-epstein-barr-reactivation\/#Top_Supplements_to_Increase_CD8_T_Cells\">CD8+ T cells<\/a>, and B cells all have <a href=\"https:\/\/selfhacked.com\/blog\/melatonin\/\">melatonin <\/a>receptors. By binding to them, melatonin can either activate or suppress these cells and modify the immune response [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC3709754\/\">R<\/a>].<\/p>\n<p>Melatonin also reduces inflammation by blocking major inflammatory hubs such as <a href=\"https:\/\/selfhacked.com\/blog\/nuclear-factor-kappa-b\/\">NF-KB<\/a>. This results in a decreased production of pro-inflammatory cytokines such as <a href=\"http:\/\/selfhacked.com\/blog\/interleukin-1\/\">IL-1\u03b2<\/a>, <a href=\"http:\/\/www.selfdecode.com\/gene\/il2\/?utm_source=seo&amp;utm_medium=selfhacked&amp;utm_campaign=id00002\">IL-2<\/a>, <a href=\"http:\/\/selfhacked.com\/blog\/interleukin-6\/\">IL-6<\/a>, <a href=\"https:\/\/www.selfdecode.com\/gene\/IL12a\/?utm_source=seo&amp;utm_medium=selfhacked&amp;utm_campaign=id00002\">IL-12<\/a>, <a href=\"https:\/\/selfdecode.com\/blog\/article\/tnf-longevity-86\">TNF<\/a>-\u03b1, and <a href=\"https:\/\/www.selfhacked.com\/blog\/interferon-gamma-how-to-increase-decrease-high-low-levels\/\">IFN-\u03b3<\/a> [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/8747536\/\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/29701146\">R<\/a>].<\/p>\n<p>Importantly, it blocks <a href=\"https:\/\/www.selfdecode.com\/gene\/NLRP3\/?utm_source=seo&amp;utm_medium=selfhacked&amp;utm_campaign=id00002\">NLRP3<\/a>. This protein complex amplifies inflammation during lung infections and contributes to the development of ARDS [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4901306\/\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC7102583\/\">R<\/a>].<\/p>\n<p>Administration of melatonin increases the cellular production of antioxidant enzymes (<a href=\"http:\/\/selfhacked.com\/blog\/the-brain-fog-gene-rs4880-or-sod2\/\">superoxide dismutase<\/a>, <a href=\"http:\/\/selfhacked.com\/blog\/30-scientifically-proven-health-benefits-glutathione\/\">glutathione<\/a><a href=\"https:\/\/www.selfdecode.com\/gene\/gpx1\/?utm_source=seo&amp;utm_medium=selfhacked&amp;utm_campaign=id00002\"> peroxidase<\/a>, and \u03b3-glutamylcysteine synthetase). Melatonin itself can bind to free radicals and render them less harmful [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC1395802\/\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2993480\/\">R<\/a>, <a href=\"https:\/\/www.sciencedirect.com\/science\/article\/abs\/pii\/S0891584999001318\">R<\/a>].<\/p>\n<p>It also blocks the generation of nitrogen radicals from nitric oxide through <a href=\"https:\/\/selfdecode.com\/blog\/article\/nos2-longevity-61\">NOS2<\/a> [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/16682846\">R<\/a>].<\/p>\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Melatonin can modify the immune response and block inflammatory signals.<\/div><\/div>\n<h3><span id=\"ARDS_and_Lung_Failure\">ARDS and Lung Failure<\/span><\/h3>\n<p>In line with this, it relieved acute lung injury and prevented its progression to ARDS in rats and mice by blocking NLRP3 [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/26888116\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/25491480\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/28370493\">R<\/a>].<\/p>\n<p>Melatonin also attenuated the response to sepsis through this mechanism [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/26681113\">R<\/a>].<\/p>\n<p>In rats with ventilator-induced injuries, a compound that activates the same receptor as melatonin (ramelteon) reduced lung inflammation and swelling by increasing the production of the anti-inflammatory cytokine <a href=\"http:\/\/selfhacked.com\/blog\/il-10\/\">IL-10<\/a> [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC7059294\/\">R<\/a>].<\/p>\n<p>In rats with pancreatitis, which may cause ARDS, melatonin relieved the decline in lung function through its anti-inflammatory and antioxidant properties [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24330221\">R<\/a>].<\/p>\n<p>In mice infected with RSV, melatonin reduced lung injury by blocking <a href=\"https:\/\/selfhacked.com\/blog\/oxidative-stress-101\/\">oxidative stress<\/a> and the inflammatory response. Similarly, it prevented the activation of the inflammatory proteins TLR3 and NF-kB in cells infected with this virus [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/20070490\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/18312297\">R<\/a>].<\/p>\n<p>In two clinical trials on almost 200 preterm babies with respiratory distress syndrome, supplementation with melatonin reduced the levels of some inflammatory and oxidative markers that seem associated with this condition (the levels of IL-6, <a href=\"http:\/\/selfhacked.com\/blog\/il-8\/\">IL-8<\/a>, <a href=\"https:\/\/selfdecode.com\/blog\/article\/tnf-thyroid-health-89\">TNF<\/a><a href=\"http:\/\/selfhacked.com\/blog\/supplements-lifestyle-factors-influence-tnf-interleukin-6-il-6\/\">-alpha<\/a>, and nitrite\/nitrate) [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/15066049\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/15168319\">R<\/a>].<\/p>\n<p>However, <strong>this condition is totally different from ARDS<\/strong> despite having a similar name and also causing labored breathing. Respiratory distress syndrome occurs in premature babies because their lungs are underdeveloped and cannot produce the foamy substance that keeps them expanded (surfactant). As a result, the lungs collapse and cannot breathe in enough oxygen [<a href=\"https:\/\/www.nhlbi.nih.gov\/health-topics\/respiratory-distress-syndrome\">R<\/a>].<\/p>\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Melatonin protected preterm babies from respiratory distress syndrome and prevented ARDS in lab animals.<\/div><\/div>\n<h3><span id=\"COPD\">COPD<\/span><\/h3>\n<p>In rats with COPD caused by cigarette smoke and bacterial lipopolysaccharide (<a href=\"https:\/\/www.selfhacked.com\/blog\/lipopolysaccharides\/\">LPS<\/a>), melatonin reduced airway inflammation [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/29990691\">R<\/a>].<\/p>\n<p>In guinea pigs exposed to air pollution, the antioxidant activity of melatonin relieved chronic cough [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/29303602\">R<\/a>].<\/p>\n<p>Melatonin also protected the lungs of rabbits exposed to cigarette smoke [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/16771911\">R<\/a>].<\/p>\n<h3><span id=\"Sarcoidosis\">Sarcoidosis<\/span><\/h3>\n<p>In a small trial on 18 people with sarcoidosis who failed to respond to conventional treatment, melatonin improved lung damage [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/16879313\">R<\/a>].<\/p>\n<h3><span id=\"Potential_Effects_Against_COVID-19\">Potential Effects Against COVID-19<\/span><\/h3>\n<p>As previously described, melatonin seems to counteract the negative effects of viral infections, including the development of ARDS, through its anti-inflammatory and antioxidant activities. Although evidence is lacking, some scientists believe it may have this protective effect in people infected with SARS-CoV-2 as well [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC7102583\/\">R<\/a>].<\/p>\n<p>Melatonin has been reported to indirectly regulate the production of the <a href=\"https:\/\/selfdecode.com\/blog\/article\/ace2-coronavirus-128\">ACE2<\/a> receptor, which the SARS-CoV-2 virus uses to enter cells. It may do so by blocking calmodulin, a protein that interacts with ACE2 to promote its expression at the cell surface [<a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC7073332\/\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/12071468\/\">R<\/a>, <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC7094239\/\">R<\/a>].<\/p>\n<div class=\"sh-summary\"><div class=\"sh-summary-content\">Melatonin is anti-inflammatory, could potentially protect against ARDS, and may regulate ACE2; these are all effects that may be helpful in COVID-19 patients.<\/div><\/div>\n\n\n<p><\/p>\n","protected":false},"excerpt":{"rendered":"<p><em>This article is for informational purposes only.
