Of course mostly starting with PI3K/Akt signalling etc.)
What’s also worth mentioning is that all this popular playing around with dopamine and serotonin neurotransmitters is beside the point and has no effect if mTOR is deactivated. And playing around with cAMP in contrast is very popular again (never any real reason, explanation deemed necessary or convincing if attempted), but it’s sort of one of the neuronal counter-players to mTOR, which is certainly no less important. (It appears there is some Dopamine 1 and 2 distribution, for example, aside from cAMP overactivity with lack of mTOR being part of many side-effects.)
One shouldn’t be led amiss by most studies’ focus on “oncogenic” effects and inhibition, and therefore shallowly conclude, without specific investigation, that one can just get rid of something and that would be “good”. Chances are that’s where a lot of problems lie…

One can find a list of all the “bad” things for anything else, including mTOR inhibition. Downright silly to be so simplistic. It is extremely important in the brain for normal function.

I am looking to replace sirolimus, mycophenalate and prednisolone required due to a liver allograft.
Three years in I have had to cease CNI’s due to the side-effects but I am also concerned about drug supply into the future. I also require insulin as genetic LADA type 1, have genetic haemochromatosis and require testosterone due to primary hypogonadism. Aromatisation is a problem. Which doesn’t help!
Not asking for a regime but would appreciate any comments you might have on a “top 3” for mTOR inhibition.

I am doing cancer research – from the angle of the newer metabolic approaches to healing and or/slowing it in its tracks. So your article is a big step in that direction and opening laymen minds that complementary strategies exist in the battle against cancer.

For too long the conservative chemo/radio treatment group of oncologists have rebuffed the public that other treatments than their own can only treat cancer.
Not any more – people like you are needed to spread the word.
I am not the either/or type of researcher as I find that some good still exists in conventional therapies,
and also much good exists in alternative therapies.
Its the die-hards on both sides which may at times obscure our search for the truth.

Also, this article would be more informative if you describe what’s known about the magnitude of TOR expression as a result of leucine or insulin. The former leads to a shorter term stimulation, and the latter causes a longer time frame for TOR expression. Eating a steak and eating a bowl of oatmeal both stimulate TOR, but the former is more profound due to its greater insulin provocation
